Anastrozole Aromatase Inhibitor Plasma Drug Concentration Genome-Wide Association Study: Functional Epistatic Interaction Between SLC38A7 and ALPPL2.
Tanda M. Dudenkov,Duan Liu,Junmei Cairns,Sandhya Devarajan,Yongxian Zhuang,James N. Ingle,Aman U. Buzdar,Mark E. Robson,Michiaki Kubo,Anthony Batzler,Poulami Barman,Gregory D. Jenkins,Erin E. Carlson,Matthew P. Goetz,Donald W. Northfelt,Alvaro Moreno-Aspitia,Zeruesenay Desta,Joel M. Reid,Krishna R. Kalari,Liewei Wang,Richard M. Weinshilboum +20 more
TL;DR: A novel gene encoding an anastrozole transporter, SLC38A7, was identified, as well as epistatic interaction between SNPs in that gene and SNPs near ALPPL2 that influenced both the expression of the transporter and anast rozole plasma concentrations.
Journal ArticleDOI
Chronic cortisol differentially impacts stem cell-derived astrocytes from major depressive disorder patients.
Kelly J. Heard,Maxim N. Shokhirev,Caroline Becronis,Callie Fredlender,Nadia Zahid,Amy T. Le,Yuan Ji,Yuan Ji,Michelle K. Skime,Timothy J. Nelson,Daniel K. Hall-Flavin,Richard M. Weinshilboum,Fred H. Gage,Krishna C. Vadodaria +13 more
TL;DR: In this paper, the authors developed an in vitro model of chronic stress using human induced pluripotent stem cell (iPSC)-derived astrocytes treated with cortisol for 7 days.
Journal ArticleDOI
Prediction of short-term antidepressant response using probabilistic graphical models with replication across multiple drugs and treatment settings
Arjun P. Athreya,Tanja Brückl,Elisabeth B. Binder,A. John Rush,A. John Rush,A. John Rush,Joanna M. Biernacka,Mark A. Frye,Drew Neavin,Michelle K. Skime,Ditlev Monrad,Ravishankar K. Iyer,Taryn L. Mayes,Madhukar H. Trivedi,Rickey E. Carter,Liewei Wang,Richard M. Weinshilboum,Paul E. Croarkin,William V. Bobo +18 more
TL;DR: In this paper, the authors used probabilistic graphical models (PGMs) to identify specific depressive symptoms and thresholds of improvement that were predictive of antidepressant response by 4 weeks for a patient to achieve remission, response, or nonresponse by 8 weeks.
Journal ArticleDOI
Machine Learning Helps Identify New Drug Mechanisms in Triple-Negative Breast Cancer
Arjun P. Athreya,Alan J. Gaglio,Junmei Cairns,Krishna R. Kalari,Richard M. Weinshilboum,Liewei Wang,Zbigniew Kalbarczyk,Ravishankar K. Iyer +7 more
TL;DR: Among the identified small set of genes associated with reduced breast cancer incidence, laboratory experiments on one of the genes, CDC42, showed that its downregulation by metformin inhibited cancer cell migration and proliferation, thus validating the ability of machine learning approaches to identify biologically relevant candidates for laboratory experiments.
Journal ArticleDOI
TCF7L2 lncRNA: a link between bipolar disorder and body mass index through glucocorticoid signaling.
Duan Liu,Thanh Thanh L. Nguyen,Huanyao Gao,Huaizhi Huang,Daniel C. Kim,Brenna Sharp,Zhenqing Ye,Jeong Heon Lee,Brandon J. Coombes,Tamas Ordog,Liewei Wang,Joanna M. Biernacka,Mark A. Frye,Richard M. Weinshilboum +13 more
TL;DR: In this article, the authors investigated the role of TCF7L2 in the central nervous system (CNS) and its possible role in the effect of body mass index (BMI) interaction.
