TL;DR: It is speculated that specific inhibition of Jak3 is not sufficient to efficiently block γC cytokine signal transduction required for strong immunosuppression, and a synthetic approach toward the rapid modification of phenyl-indolyl maleimides and the discovery of potent Jak3 inhibitor 1 with high selectivity within the Jak kinase family is described.

A natural ligand for the orphan receptor GPR15 modulates lymphocyte recruitment to epithelia

TL;DR: The purification from porcine colonic tissue extracts of an agonistic ligand for GPR15 and its functional characterization identifies a receptor-ligand pair that is required for immune homeostasis at epithelia and whose modulation may represent an alternative approach to treating conditions affecting the skin such as psoriasis.
