Oxysterols direct immune cell migration via EBI2
Sébastien Hannedouche,Juan Zhang,Tangsheng Yi,Weijun Shen,Weijun Shen,Deborah Nguyen,João Pereira,Danilo Guerini,Birgit Baumgarten,Silvio Roggo,Ben Wen,Richard Knochenmuss,Sophie Noël,François Gessier,Lisa M. Kelly,Miroslava Vanek,Stephane Laurent,Inga Preuss,Charlotte Miault,Isabelle Christen,Ratnaningrum Karuna,Wei Li,Dong-In Koo,Thomas Suply,Christian Schmedt,Eric C. Peters,Rocco Falchetto,Andreas Katopodis,Carsten Spanka,Marie-Odile Roy,Michel Detheux,Yu Chen,Peter G. Schultz,Charles Y. Cho,Klaus Seuwen,Jason G. Cyster,Andreas W. Sailer +36 more
TL;DR: The identification of 7α,25-dihydroxycholesterol as a potent and selective agonist of EBI2 and its role in the adaptive immune response is described.
Journal ArticleDOI
Absolute quantification of monoclonal antibodies in biofluids by liquid chromatography-tandem mass spectrometry.
TL;DR: A lowest limit of quantification of 2 microg/mL HmAb was reached in both human and cynomolgus monkey samples, and the dynamic range of the method was 3 orders of magnitude.
Journal ArticleDOI
SpeedScreen: label-free liquid chromatography-mass spectrometry-based high-throughput screening for the discovery of orphan protein ligands.
TL;DR: A high-throughput screening methodology tailored to the discovery of ligands for known and orphan proteins is presented, as the ligands are affinity selected by incubation of the protein with mixtures of compounds in aqueous binding buffer.
Journal ArticleDOI
Identification of a Potent Janus Kinase 3 Inhibitor with High Selectivity within the Janus Kinase Family.
Gebhard Thoma,Nuninger Francois,Rocco Falchetto,Erwin Hermes,Gisele A. Tavares,Eric Vangrevelinghe,Hans-Günter Zerwes +6 more
TL;DR: It is speculated that specific inhibition of Jak3 is not sufficient to efficiently block γC cytokine signal transduction required for strong immunosuppression, and a synthetic approach toward the rapid modification of phenyl-indolyl maleimides and the discovery of potent Jak3 inhibitor 1 with high selectivity within the Jak kinase family is described.
Journal ArticleDOI
A natural ligand for the orphan receptor GPR15 modulates lymphocyte recruitment to epithelia
Thomas Suply,Sébastien Hannedouche,Nathalie Carte,Jianping Li,Bianka L. Grosshans,Michael Schaefer,Layla Raad,Valerie Beck,Solange Vidal,Agnès Hiou-Feige,Noemie Beluch,Samuel Barbieri,Johann Wirsching,Nadine Lageyre,Frank Hillger,Corinne Debon,Janet Dawson,Philip Smith,Vincent Lannoy,Michel Detheux,Francis Bitsch,Rocco Falchetto,Tewis Bouwmeester,Jeffrey A. Porter,Birgit Baumgarten,Keith Mansfield,José M. Carballido,Klaus Seuwen,Frederic Bassilana +28 more
TL;DR: The purification from porcine colonic tissue extracts of an agonistic ligand for GPR15 and its functional characterization identifies a receptor-ligand pair that is required for immune homeostasis at epithelia and whose modulation may represent an alternative approach to treating conditions affecting the skin such as psoriasis.
