Lower TCR repertoire diversity in Traj18-deficient mice.
Romain Bedel,Jennifer L. Matsuda,Manfred Brigl,Janice White,Janice White,John W. Kappler,Philippa Marrack,Laurent Gapin +7 more
TL;DR: Natural killer T cells (NKT cells) constitute a distinct subset of T lymphocytes that can modulate immune responses through the rapid release of cytokines and direct interactions with other cells of the immune system and are promising targets for immunotherapy.
Journal ArticleDOI
TCR signal strength controls thymic differentiation of iNKT cell subsets
Kathryn D. Tuttle,S. Harsha Krovi,Jingjing Zhang,Romain Bedel,Romain Bedel,Laura Harmacek,Lisa K. Peterson,Lisa K. Peterson,Leonard L. Dragone,Adam R. Lefferts,Catherine Halluszczak,Kent Riemondy,Jay R. Hesselberth,Anjana Rao,Anjana Rao,Brian P. O'Connor,Philippa Marrack,James P. Scott-Browne,Laurent Gapin +18 more
TL;DR: It is shown, by analysing mice carrying mutant Zap70 genes, that T cell receptor signaling strength induces epigenetic changes of genes to modulate iNKT lineages, suggesting that variable TCR signaling modulates regulatory element activity at NFAT and Egr binding sites exerting a determinative influence on the dynamics of gene enhancer accessibility and the developmental fate of iN KT cells.
Journal ArticleDOI
Vβ2 natural killer T cell antigen receptor-mediated recognition of CD1d-glycolipid antigen
Onisha Patel,Daniel G. Pellicci,Adam P Uldrich,Lucy C. Sullivan,Mugdha Bhati,Melissa A McKnight,Stewart K. Richardson,Amy R. Howell,Thierry Mallevaey,Jingjing Zhang,Romain Bedel,Gurdyal S. Besra,Andrew G. Brooks,Lars Kjer-Nielsen,James McCluskey,Steven A. Porcelli,Laurent Gapin,Jamie Rossjohn,Jamie Rossjohn,Dale I. Godfrey +19 more
TL;DR: It is shown that there is a greater interplay between the germline– and nongermline–encoded loops within the TCR β-chain of the Vβ2 NKT TCR that enables CD1d-Ag ligation.
Journal ArticleDOI
Effective functional maturation of invariant natural killer T cells is constrained by negative selection and T-cell antigen receptor affinity
Romain Bedel,Richard Berry,Thierry Mallevaey,Jennifer L. Matsuda,Jingjing Zhang,Dale I. Godfrey,Jamie Rossjohn,Jamie Rossjohn,John W. Kappler,Philippa Marrack,Laurent Gapin +10 more
TL;DR: Proper development of fully functional iNKT cells is constrained by a limited range of TCR affinity that plays a key role in triggering the iN KT cell-differentiation pathway.
Journal ArticleDOI
Mutation of the Traj18 gene segment using TALENs to generate Natural Killer T cell deficient mice
Jingjing Zhang,Romain Bedel,Romain Bedel,S. Harsha Krovi,Kathryn D. Tuttle,Bicheng Zhang,James Gross,Laurent Gapin,Jennifer L. Matsuda +8 more
TL;DR: A new NKT-deficient mouse strain is created using transcription activator-like effector nuclease (TALEN) technology to only disrupt the expression of Jα18, leaving the remaining Jα repertoire unperturbed and serving as a new model of iNKT cell deficiency.
