Journal ArticleDOI
Iron Oxide Nanoparticles Induce Dopaminergic Damage: In vitro Pathways and In Vivo Imaging Reveals Mechanism of Neuronal Damage
Syed Z. Imam,Susan M. Lantz-McPeak,Elvis Cuevas,Hector Rosas-Hernandez,Serguei Liachenko,Yongbin Zhang,Sumit Sarkar,Jaivijay Ramu,Bonnie L. Robinson,Yvonne Jones,Bobby Gough,Merle G. Paule,Syed F. Ali,Zbigniew Binienda +13 more
TL;DR: The data demonstrate a neurotoxic potential of very small size iron nanoparticles and suggest that use of these ferric oxide nanoparticles may result in neurotoxicity, thereby limiting their clinical application.
Journal ArticleDOI
The Pathophysiological Sequence of Glucocorticoid-Induced Osteonecrosis of the Femoral Head in Male Mice
Robert S. Weinstein,Erin A. Hogan,Michael J. Borrelli,Serguei Liachenko,Charles A. O'Brien,Stavros C. Manolagas +5 more
TL;DR: The results indicate that the Femoral head is a particularly sensitive anatomical site to the adverse effects of glucocorticoid excess on bone and that decreases of Hif-1α and VEGF expression, bone vascularity, and strength precede the loss of bone mass and microarchitectural deterioration, thus rendering the femoral head vulnerable to collapse.
Journal ArticleDOI
Translational Biomarkers of Neurotoxicity: A Health and Environmental Sciences Institute Perspective on the Way Forward.
Ruth Roberts,Michael Aschner,David O. Calligaro,Tomás R. Guilarte,Joseph P. Hanig,David W. Herr,Thomas J. Hudzik,Andreas Jeromin,Mary Jeanne Kallman,Serguei Liachenko,James J. Lynch,Diane B. Miller,Virginia C. Moser,James P. O'Callaghan,William Slikker,Merle G. Paule +15 more
TL;DR: The HESI framework is utilized to propose a consensus on the relative potential of currently described biomarkers of neurotoxicity to assess utility of the selected biomarkers using a nonclinical model.
Journal ArticleDOI
The use of MRI to assist the section selections for classical pathology assessment of neurotoxicity.
Joseph P. Hanig,Merle G. Paule,Jaivijay Ramu,Larry C. Schmued,Tetyana Konak,Srinivasulu Chigurupati,William Slikker,Sumit Sarkar,Serguei Liachenko +8 more
TL;DR: The application of full brain MRI imaging that informs neuropathology offers the potential to dramatically improve detection of neurotoxicity produced by new drugs and facilitate new drug development, review and approval processes, and to qualify an imaging biomarker of neuro Pathology.
Journal ArticleDOI
A multi-center preclinical study of gadoxetate DCE-MRI in rats as a biomarker of drug induced inhibition of liver transporter function.
Anastassia Karageorgis,Stephen C. Lenhard,Brittany Yerby,Mikael Forsgren,Serguei Liachenko,Edvin Johansson,Mark Pilling,Richard A. Peterson,Xi Yang,Dominic P. Williams,Sharon E. Ungersma,Ryan E. Morgan,Kim L. R. Brouwer,Beat M. Jucker,Paul D. Hockings +14 more
TL;DR: Rate constants of gadoxetate uptake and excretion are sensitive and robust biomarkers to detect early changes in hepatobiliary transporter function in vivo in rats prior to established biomarkers of liver toxicity.
