Journal ArticleDOI
Mutual protection of ribosomal proteins L5 and L11 from degradation is essential for p53 activation upon ribosomal biogenesis stress
Sladana Bursac,Maja Cokarić Brdovčak,Martin Pfannkuchen,Ines Orsolic,Lior Golomb,Yan Zhu,Chen Katz,Lilyn Daftuar,Kristina Grabušić,Iva Vukelić,Vedrana Filić,Moshe Oren,Carol Prives,Siniša Volarević +13 more
TL;DR: It is reported that the disrupted nucleoli may provide a platform for L5- and L11-dependent p53 activation, implying a role for the nucleolus in p53activation by ribosomal biogenesis stress.
Journal ArticleDOI
Nucleolus as an emerging hub in maintenance of genome stability and cancer pathogenesis.
TL;DR: The focus will be on features of rDNA genes, which make them highly vulnerable to DNA damage and intra- and interchromosomal recombination as well as built-in mechanisms that prevent and repair rDNA damage, and how dysregulation of this interplay affects genome-wide DNA stability, gene expression and the balance between euchromatin and heterochromatin.
Journal ArticleDOI
Activation of the tumor suppressor p53 upon impairment of ribosome biogenesis
TL;DR: Recent advances made toward understanding of this newly-recognized checkpoint signaling pathway, its role in health and disease, and possible future directions in this exciting research field are reviewed.
Journal ArticleDOI
Functional interplay between the DNA-damage-response kinase ATM and ARF tumour suppressor protein in human cancer
Georgia Velimezi,Michalis Liontos,Konstantinos Vougas,Theodoros I. Roumeliotis,Theodoros I. Roumeliotis,Jirina Bartkova,Maria Sideridou,Ayguel Dereli-Oz,Maciej K. Kocylowski,Ioannis S. Pateras,Kostas Evangelou,Athanassios Kotsinas,Ines Orsolic,Sladana Bursac,Maja Cokaric-Brdovcak,Vassilis Zoumpourlis,Dimitris Kletsas,George Papafotiou,Apostolos Klinakis,Siniša Volarević,Wei Gu,Jiri Bartek,Thanos D. Halazonetis,Vassilis G. Gorgoulis,Vassilis G. Gorgoulis +24 more
TL;DR: In human oncogene-transformed and cancer cells, ATM suppressed ARF protein levels and activity in a transcription-independent manner and inhibition of ATM stimulated the tumour-suppressive effects of ARF.
Journal ArticleDOI
RNA-interference screen for p53 regulators unveils a role of WDR75 in ribosome biogenesis.
TL;DR: In this paper, a high-content siRNA-based screen of 175 human ribosome biogenesis factors, searching for impact on p53, was performed and it was shown that WDR75 is required for pre-rRNA transcription through supporting the maintenance of physiological levels of RPA194, a key subunit of the RNA polymerase I.
