Abstract: Altruistic behavior varies considerably across people and decision contexts. The relevant computational and motivational mechanisms that underlie its heterogeneity, however, are poorly understood. Using a charitable giving task together with multivariate decoding techniques, we identified three distinct psychological mechanisms underlying altruistic decision-making (empathy, perspective taking, and attentional reorienting) and linked them to dissociable neural computations. Neural responses in the anterior insula (AI) (but not temporoparietal junction [TPJ]) encoded trial-wise empathy for beneficiaries, whereas the TPJ (but not AI) predicted the degree of perspective taking. Importantly, the relative influence of both socio-cognitive processes differed across individuals: participants whose donation behavior was heavily influenced by affective empathy exhibited higher predictive accuracies for generosity in AI, whereas those who strongly relied on cognitive perspective taking showed improved predictions of generous donations in TPJ. Furthermore, subject-specific contributions of both processes for donations were reflected in participants' empathy and perspective taking responses in a separate fMRI task (EmpaToM), suggesting that process-specific inputs into altruistic choices may reflect participants' general propensity to either empathize or mentalize. Finally, using independent attention task data, we identified shared neural codes for attentional reorienting and generous donations in the posterior superior temporal sulcus, suggesting that domain-general attention shifts also contribute to generous behavior (but not in TPJ or AI). Overall, our findings demonstrate highly specific roles of AI for affective empathy and TPJ for cognitive perspective taking as precursors of prosocial behavior and suggest that these discrete routes of social cognition differentially drive intraindividual and interindividual differences in altruistic behavior.

Abstract: The anterior insula (AI) and mid-anterior cingulate cortex (mACC) have repeatedly been implicated in first-hand and vicarious experiences of pain, disgust and unfairness. However, it is debated whether these regions process different aversive events through a common modality-independent code, reflecting the shared unpleasantness of the experiences or through independent modality-specific representations. Using functional magnetic resonance imaging, we subjected 19 participants (and 19 confederates) to equally unpleasant painful and disgusting stimulations, as well as unfair monetary treatments. Multivoxel pattern analysis identified modality-independent activation maps in the left AI and mACC, pointing to common coding of affective unpleasantness, but also response patterns specific for the events' sensory properties and the person to whom it was addressed, particularly in the right AI. Our results provide evidence of both functional specialization and integration within AI and mACC, and support a comprehensive role of this network in processing aversive experiences for self and others.

Abstract: Although the processes that underlie sharing others' emotions (empathy) and understanding others' mental states (mentalizing, Theory of Mind) have received increasing attention, it is yet unclear how they relate to each other. For instance, are people who strongly empathize with others also more proficient in mentalizing? And (how) do the neural networks supporting empathy and mentalizing interact? Assessing both functions simultaneously in a large sample (N = 178), we show that people's capacities to empathize and mentalize are independent, both on a behavioral and neural level. Thus, strong empathizers are not necessarily proficient mentalizers, arguing against a general capacity of social understanding. Second, we applied dynamic causal modeling to investigate how the neural networks underlying empathy and mentalizing are orchestrated in naturalistic social settings. Results reveal that in highly emotional situations, empathic sharing can inhibit mentalizing-related activity and thereby harm mentalizing performance. Taken together, our findings speak against a unitary construct of social understanding and suggest flexible interplay of distinct social functions.

Abstract: Prosocial behavior is crucial for functioning societies. However, its reliable scientific assessment and the understanding of its underlying structure are still a challenge. We integrated 14 paradigms from diverse disciplines to identify reliable and method-independent subcomponents of human prosociality; 329 participants performed game theoretical paradigms and hypothetical distribution tasks commonly used in behavioral economics and completed interactive computer tasks and self-reports typically employed in psychology. Four subcomponents of prosociality were identified by exploratory factor analysis and verified by confirmatory factor analysis in an independent sample: altruistically motivated prosocial behavior, norm motivated prosocial behavior, strategically motivated prosocial behavior, and self-reported prosocial behavior. Altruistically motivated behavior was related to gender, to enhanced cognitive skills, and to reduced negative affect. Our study provides a crucial step toward an overarching framework on prosocial behavior that will benefit future research on predictors, neural underpinnings, and plasticity of human cooperation and prosociality.

Abstract: Functional neuroimaging studies have suggested the existence of 2 largely distinct social cognition networks, one for theory of mind (taking others' cognitive perspective) and another for empathy (sharing others' affective states). To address whether these networks can also be dissociated at the level of brain structure, we combined behavioral phenotyping across multiple socio-cognitive tasks with 3-Tesla MRI cortical thickness and structural covariance analysis in 270 healthy adults, recruited across 2 sites. Regional thickness mapping only provided partial support for divergent substrates, highlighting that individual differences in empathy relate to left insular-opercular thickness while no correlation between thickness and mentalizing scores was found. Conversely, structural covariance analysis showed clearly divergent network modulations by socio-cognitive and -affective phenotypes. Specifically, individual differences in theory of mind related to structural integration between temporo-parietal and dorsomedial prefrontal regions while empathy modulated the strength of dorsal anterior insula networks. Findings were robust across both recruitment sites, suggesting generalizability. At the level of structural network embedding, our study provides a double dissociation between empathy and mentalizing. Moreover, our findings suggest that structural substrates of higher-order social cognition are reflected rather in interregional networks than in the the local anatomical markup of specific regions per se.

Abstract: Autism spectrum disorder (ASD) shows deficits in self-other distinction during theory of mind (ToM). Here we investigated whether ASD patients also show difficulties in self-other distinction during empathy and if potential deficits are linked to dysfunctional resting-state connectivity patterns. In a first study, ASD patients and controls performed an emotional egocentricity paradigm and a ToMtask. In the second study, resting-state connectivity of right temporo-parietal junction and right supramarginal gyrus(rSMG) were analysed using a large-scale fMRI data set. ASD patients exhibited deficient ToM but normal emotional egocentricity, which was paralleled by reduced connectivity of regions of the ToM network and unimpaired rSMG network connectivity. These results suggest spared self-other distinction during empathy and an intact rSMG network in ASD.

Abstract: Human civilization is based on the successful pursuit of long-term goals, requiring the ability to forego immediate pleasure for the sake of larger future rewards This ability improves with age, but the precise cognitive and neural mechanisms underlying its development remain elusive The developmental changes could result either from younger children valuing immediate rewards more strongly or because older children become better at controlling their impulses By implementing 2 tasks, a choice-independent valuation task and an intertemporal choice task, both behaviorally and using fMRI in twenty 6- to 13-year old children, we show developmental improvements in behavioral control to uniquely account for age-related changes in temporal discounting We show further that overcoming temptation during childhood occurs as a function of an age-related increase in functional coupling between value signals in the ventromedial prefrontal cortex and brain regions dedicated to behavioral control, such as left dorsolateral prefrontal cortex during choice These findings can help to devise measures that reduce the substantial costs of impatience to society

Abstract: Emotion regulation in the ongoing presence of a threat is essential for adaptive behavior. Threatening situations change over time and, as a consequence, require a fine-tuned, dynamic regulation of arousal to match the current state of the environment. Constructs such as cognitive flexibility, heart rate variability, and resilience have been proposed as resources for adaptive emotion regulation, especially in a moment-to-moment fashion. Nevertheless, none of these constructs has been empirically related to the dynamic regulation of arousal as it unfolds over the course of a prolonged threatening episode. Here, we do so by placing participants in a threatening and evolving immersive virtual environment called Room 101, while recording their skin conductance. Subsequently, participants rated their subjective arousal continuously over the course of the experience. Participants who had shown greater cognitive flexibility in a separate task (i.e., fewer task-switching costs when switching to evaluating the valence of positive stimuli) showed better regulation of physiological arousal (skin conductance level), during less-threatening phases of Room 101. Individuals with higher trait resilience and individuals with higher resting heart rate variability showed more regulation in terms of their subjective arousal experience. The results indicate that emotional, cognitive, and physiological flexibility support nuanced adaptive regulation of objective and experienced arousal in the ongoing presence of threats. Furthermore, the results indicate that these forms of flexibility differentially affect automatic and objective versus reflective and subjective processes.

Abstract: One important aspect of metacognition is the ability to accurately evaluate one's performance. People vary widely in their metacognitive ability and in general are too confident when evaluating their performance. This often leads to poor decision making with potentially disastrous consequences. To further our understanding of the neural underpinnings of these processes, this fMRI study investigated inter-individual differences in metacognitive ability and effects of trial-by-trial variation in subjective feelings of confidence when making metacognitive assessments. Participants (N = 308) evaluated their performance in a high-level social and cognitive reasoning task. The results showed that higher metacognitive accuracy was associated with a decrease in activation in the anterior medial prefrontal cortex, an area previously linked to metacognition on perception and memory. Moreover, the feeling of confidence about one's choices was associated with an increase of activation in reward, memory and motor related areas including bilateral striatum and hippocampus, while less confidence was associated with activation in areas linked with negative affect and uncertainty, including dorsomedial prefrontal and bilateral orbitofrontal cortex. This might indicate that positive affect is related to higher confidence thereby biasing metacognitive decisions towards overconfidence. In support, behavioural analyses revealed that increased confidence was associated with lower metacognitive accuracy.

Abstract: Humans have the ability to reflect upon their perception, thoughts, and actions, known as metacognition (MC). The brain basis of MC is incompletely understood, and it is debated whether MC on different processes is subserved by common or divergent networks. We combined behavioral phenotyping with multi-modal neuroimaging to investigate whether structural substrates of individual differences in MC on higher-order cognition (MC-C) are dissociable from those underlying MC on perceptual accuracy (MC-P). Motivated by conceptual work suggesting a link between MC and cognitive perspective taking, we furthermore tested for overlaps between MC substrates and mentalizing networks. In a large sample of healthy adults, individual differences in MC-C and MC-P did not correlate. MRI-based cortical thickness mapping revealed a structural basis of this independence, by showing that individual differences in MC-P related to right prefrontal cortical thickness, while MC-C scores correlated with measures in lateral prefrontal, temporo-parietal, and posterior midline regions. Surface-based superficial white matter diffusivity analysis revealed substrates resembling those seen for cortical thickness, confirming the divergence of both MC faculties using an independent imaging marker. Despite their specificity, substrates of MC-C and MC-P fell clearly within networks known to participate in mentalizing, confirmed by task-based fMRI in the same subjects, previous meta-analytical findings, and ad-hoc Neurosynth-based meta-analyses. Our integrative multi-method approach indicates domain-specific substrates of MC; despite their divergence, these nevertheless likely rely on component processes mediated by circuits also involved in mentalizing. Abstract: A classical model of human attention holds that independent neural networks realize stimulus-driven reorienting and executive control of attention. Questioning full independence, the two functions do, however, engage overlapping networks with activations in cingulo-opercular regions such as anterior insula (AI) and a reverse pattern of activation (stimulus-driven reorienting), and deactivation (executive control) in temporoparietal junction (TPJ). To test for independent versus shared neural mechanisms underlying stimulus-driven and executive control of attention, we used fMRI and a task that isolates individual from concurrent demands in both functions. Results revealed super-additive increases of left AI activity and behavioral response costs under concurrent demands, suggesting a common bottleneck for stimulus-driven reorienting and executive control of attention. These increases were mirrored by non-additive decreases of activity in the default mode network (DMN), including posterior TPJ, regions where activity increased with off-task processes. The deactivations in posterior TPJ were spatially separated from stimulus-driven reorienting related activation in anterior TPJ, a differentiation that replicated in task-free resting state. Furthermore, functional connectivity indicated inhibitory coupling between posterior TPJ and AI during concurrent attention demands. These results demonstrate a role of AI in stimulus-driven and executive control of attention that involves down-regulation of internally directed processes in DMN.

Abstract: This paper examines the reflexive interplay between individual decisions and social forces to analyze the evolution of cooperation in the presence of "multi-directedness", whereby people's preferences depend on their psychological motives. People have access to multiple, discrete motives. Different motives may be activated by different social settings. Inter-individual differences in dispositional types affect the responsiveness of people's motives to their social settings. The evolution of these dispositional types is driven by changes in the frequencies of social settings. In this context, economic policies can influence economic decisions not merely by modifying incentives operating through given preferences, but also by influencing people's motives (thereby changing their preferences) and by changing the distribution of dispositional types in the population (thereby changing their motivational responsiveness to social settings).

Abstract: Diagnostic criteria for borderline personality disorder (BPD) include instability in identity and interpersonal relationships. Here, we probed whether instability is already present in BPD patients' thoughts about themselves and others. We tested BPD patients (N=27) and healthy controls (N=25) with a mind-wandering task that assesses content and variability of stimulus-independent self-generated thoughts. Multi-level modeling revealed that while BPD patients and healthy controls mind-wander to a similar extent, BPD patients' thoughts are colored predominantly negatively. Most importantly, although their thoughts concerned the self and others as much as in controls, they fluctuated more strongly in the degree to which their thoughts concerned themselves and others and also gave more extreme ratings. Self- and other related thoughts that were more extreme were also more negative in valence. The increased variability supports current conceptualizations of BPD and may account for the instability in identity and interpersonal relationships.

Abstract: Autism spectrum disorders are neurodevelopmental conditions with severe impairments in social communication and interaction. Pioneering research suggests that oxytocin can improve motivation, cognition and attention to social cues in patients with autism spectrum disorder. The aim of this clinical trial is to characterize basic mechanisms of action of acute oxytocin treatment on neural levels and to relate these to changes in different levels of socio-affective and -cognitive functioning. This clinical study is a randomized, double-blind, cross-over, placebo-controlled, multicenter functional magnetic resonance imaging study with two arms. A sample of 102 male autism spectrum disorder patients, diagnosed with Infantile Autistic Disorder (F84.0 according to ICD-10), Asperger Syndrome (F84.5 according to ICD-10), or Atypical Autism (F84.1 according to ICD-10) will be recruited and will receive oxytocin and placebo nasal spray on two different days. Autism spectrum disorder patients will be randomized to determine who receives oxytocin on the first and who on the second visit. Healthy control participants will be recruited and case-control matched to the autism spectrum disorder patients. The primary outcome will be neural network activity, measured with functional magnetic resonance imaging while participants perform socio-affective and -cognitive tasks. Behavioral markers such as theory of mind accuracy ratings and response times will be assessed as secondary outcomes in addition to physiological measures such as skin conductance. Trait measures for alexithymia, interpersonal reactivity, and social anxiety will also be evaluated. Additionally, we will analyze the effect of oxytocin receptor gene variants and how these potentially influence the primary and secondary outcome measures. Functional magnetic resonance imaging assessments will take place at two time points which will be scheduled at least two weeks apart to ensure a sufficient wash-out time after oxytocin treatment. The study has been approved by an ethical review board and the competent authority. Revealing the mechanisms of acute oxytocin administration, especially on the socio-affective and -cognitive domains at hand, will be a further step towards novel therapeutic interventions regarding autism. Abstract: Pain features centrally in numerous illnesses and generates enormous public health costs. Despite its ubiquity, the psychological and neurophysiological nature of pain remains controversial. Here, we survey one controversy in particular: the relation between nociceptive pain, which is somatic in origin, and empathic pain, which arises from observing others in pain. First, we review evidence for neural overlap between nociceptive and empathic pain and what this overlap implies about underlying mental representations. Then, we propose a framework for understanding the nature of the psychological and neurophysiological correspondence across these types of ‘pain’. This framework suggests new directions for research that can better identify shared and dissociable representations underlying different types of distress, and can inform theories about the nature of pain.
