Journal ArticleDOI
Obesity accelerates epigenetic aging of human liver
Steve Horvath,Wiebke Erhart,Mario Brosch,Ole Ammerpohl,Witigo von Schönfels,M Ahrens,Nils Heits,Jordana T. Bell,Pei-Chien Tsai,Tim D. Spector,Panos Deloukas,Reiner Siebert,Bence Sipos,Thomas Becker,Christoph Röcken,Clemens Schafmayer,Jochen Hampe +16 more
TL;DR: The epigenetic age acceleration, which is not reversible in the short term after rapid weight loss induced by bariatric surgery, may play a role in liver-related comorbidities of obesity, such as insulin resistance and liver cancer.
Journal ArticleDOI
Genomic regulatory blocks encompass multiple neighboring genes and maintain conserved synteny in vertebrates.
Hiroshi Kikuta,Mary Laplante,Pavla Navratilova,Anna Z. Komisarczuk,Pär G. Engström,David Fredman,Altuna Akalin,Mario Caccamo,Ian M Sealy,Kerstin Howe,Julien Ghislain,Guillaume Pézeron,Philippe Mourrain,Staale Ellingsen,Andrew C. Oates,Christine Thisse,Bernard Thisse,Isabelle Foucher,Birgit Adolf,Andrea Geling,Boris Lenhard,Thomas Becker +21 more
TL;DR: It is shown that loss of bystander genes and other mutational events suffered by duplicated GRBs in teleost genomes permits target gene identification and HCNE/target gene assignment, which explains the absence of evolutionary breakpoints from large vertebrate chromosomal segments and will aid in the recognition of position effect mutations within human GRBs.
Journal ArticleDOI
Insertional mutagenesis and rapid cloning of essential genes in zebrafish
Nicholas Gaiano,Adam Amsterdam,Koichi Kawakami,Miguel L. Allende,Thomas Becker,Nancy Hopkins +5 more
TL;DR: To facilitate the cloning of vertebrate genes that are important during embryogenesis, an insertional mutagenesis strategy in zebrafish using a retroviral vector is developed and three insertional mutants with embryonic lethal phenotypes are obtained.
Journal ArticleDOI
Cryo-EM structure and rRNA model of a translating eukaryotic 80S ribosome at 5.5-A resolution.
Jean Paul Armache,Alexander Jarasch,Andreas M. Anger,Elizabeth Villa,Thomas Becker,Shashi Bhushan,Fabrice Jossinet,Michael Habeck,Gülcin Dindar,Sibylle Franckenberg,Viter Márquez,Thorsten Mielke,Michael Thomm,Otto Berninghausen,B. Beatrix,Johannes Söding,Eric Westhof,Daniel N. Wilson,Roland Beckmann +18 more
TL;DR: Accurate assignment of the rRNA expansion segments (ES) and variable regions has revealed unique ES–ES and r-protein–ES interactions, providing insight into the structure and evolution of the eukaryotic ribosome.
Journal ArticleDOI
Genomic regulatory blocks underlie extensive microsynteny conservation in insects
TL;DR: Evidence that microsynteny has been retained to keep large arrays of highly conserved noncoding elements (HCNEs) intact is presented and differences in core promoters between the target genes and the unrelated genes that offer an explanation for the differences in their responsiveness to enhancers are described.
