Journal ArticleDOI
Essential role for the p110δ isoform in phosphoinositide 3-kinase activation and cell proliferation in acute myeloid leukemia
Pierre Sujobert,Valérie Bardet,Pascale Cornillet-Lefebvre,Joel S. Hayflick,Nolwen Prie,Frederic Verdier,Bart Vanhaesebroeck,Odile Muller,Florence Pesce,Norbert Ifrah,Mathilde Hunault-Berger,Christian Berthou,Bruno Villemagne,Eric Jourdan,Bruno Audhuy,Eric Solary,Brigitte Witz,Jean Luc Harousseau,Chantal Himberlin,Thierry Lamy,Bruno Lioure,Jean-Yves Cahn,François Dreyfus,Patrick Mayeux,Catherine Lacombe,Didier Bouscary +25 more
TL;DR: It is shown that the p110δ isoform of PI3K is consistently expressed at a high level in blast cells from AML, in contrast to the other class I isoforms, the expression of which was very variable among patients.
Journal ArticleDOI
Predictive factors of response and survival in myelodysplastic syndrome treated with erythropoietin and G-CSF: the GFM experience.
Sophie Park,Sophie Grabar,Charikleia Kelaidi,Odile Beyne-Rauzy,Françoise Picard,Valérie Bardet,Valérie Coiteux,Geneviève Leroux,Pascale Lepelley,Marie-Thérèse Daniel,Stéphane Cheze,Beatrice Mahe,Augustin Ferrant,Christophe Ravoet,Martine Escoffre-Barbe,Lionel Ades,Norbert Vey,L. Aljassem,Aspasia Stamatoullas,Lionel Mannone,Hervé Dombret,Keith Bourgeois,Peter L. Greenberg,Pierre Fenaux,François Dreyfus +24 more
TL;DR: Multivariate adjusted comparisons of survival between this cohort and the untreated MDS cohort used to design IPSS showed similar rate of progression to acute myeloid leukemia in both cohorts, but significantly better overall survival in this cohort, suggesting that epoetin or DAR treatment may have a favorable survival impact in MDS.
Journal ArticleDOI
Mammalian target of rapamycin (mTOR) inhibition activates phosphatidylinositol 3-kinase/Akt by up-regulating insulin-like growth factor-1 receptor signaling in acute myeloid leukemia: rationale for therapeutic inhibition of both pathways
Jerome Tamburini,Nicolas Chapuis,Valérie Bardet,Sophie Park,Pierre Sujobert,Lise Willems,Lise Willems,Lise Willems,Norbert Ifrah,François Dreyfus,Patrick Mayeux,Patrick Mayeux,Patrick Mayeux,Catherine Lacombe,Didier Bouscary +14 more
TL;DR: The results suggest that dual inhibition of the mTORC1 complex and the IGF-1/IGF-1R/PI3K/Akt pathway in AML may enhance the efficacy of mTOR inhibitors in treatment of this disease.
Journal ArticleDOI
Role of the PI3K/AKT and mTOR signaling pathways in acute myeloid leukemia
Sophie Park,Nicolas Chapuis,Jerome Tamburini,Valérie Bardet,Pascale Cornillet-Lefebvre,Lise Willems,Alexa S. Green,Patrick Mayeux,Catherine Lacombe,Didier Bouscary +9 more
TL;DR: The results open the way for the design of direct inhibitors of protein synthesis as novel acute myeloid leukemia therapies and also for the development of second generation mTOR inhibitors (the TORKinhibs).
Journal ArticleDOI
Inhibiting glutamine uptake represents an attractive new strategy for treating acute myeloid leukemia.
Lise Willems,Lise Willems,Nathalie Jacque,Arnaud Jacquel,Nathalie Neveux,Thiago Trovati Maciel,Mireille Lambert,Alain Schmitt,Laury Poulain,Alexa S. Green,Madalina Uzunov,Olivier Kosmider,Olivier Kosmider,Isabelle Radford-Weiss,Ivan C. Moura,Patrick Auberger,Norbert Ifrah,Valérie Bardet,Valérie Bardet,Nicolas Chapuis,Nicolas Chapuis,Catherine Lacombe,Catherine Lacombe,Patrick Mayeux,Patrick Mayeux,Jerome Tamburini,Jerome Tamburini,Didier Bouscary,Didier Bouscary +28 more
TL;DR: It is shown that glutamine removal inhibits mTORC1 and induces apoptosis in AML cells, and that l-ases upregulate glutamine synthase expression in leukemic cells and that a GS knockdown enhances l-ase-induced apoptotic response in someAML cells.
