Endoplasmic Reticulum Stress Regulators: New Drug Targets for Parkinson’s Disease
Vera Kovaleva,Mart Saarma +1 more
TL;DR: In this article, the authors discuss ER stress as a therapeutic target to treat Parkinson's disease and summarize the existing preclinical data on the regulation of ER stress for PD treatment; they point out the crucial aspects for successful clinical translation of UPR-regulating GFs and new prospective in GFs-based treatments of PD, focusing on ER stress regulation.
Journal ArticleDOI
MANF regulates neuronal survival and UPR through its ER-located receptor IRE1α.
Vera Kovaleva,Li-Ying Yu,Larisa Yur'yevna Ivanova,Olesya G. Shpironok,J. Nam,Ave Eesmaa,Esa-Pekka Kumpula,Sven Sakson,Urve Toots,Mart Ustav,Juha T. Huiskonen,Merja H. Voutilainen,Päivi Lindholm,Mati Karelson,Mart Saarma +14 more
TL;DR: In this paper , the binding interface between MANF and IRE1α was identified, and the expression of wild-type MANF, but not its IRE 1α binding-deficient mutant, attenuates UPR signaling by decreasing IREα oligomerization; phosphorylation; splicing of Xbp1, Atf6, and Txnip levels; and protecting neurons from ER stress-induced death.

Antidepressants bind to cholesterol-interaction motif of TRKB neurotrophin receptor
Plinio C. Casarotto,Mykhailo Girych,Senem Merve Fred,Rafael Moliner,Giray Enkavi,Caroline Biojone,Cecilia Cannarozzo,Cecilia A. Brunello,Anna Steinzeig,Frederike Winkel,Sudarshan Patil,Stefan Vestring,Tsvetan Serchov,Vera Kovaleva,Cassiano Raf Diniz,Cassiano Raf Diniz,Liina Laukkanen,Iseline Cardon,Hanna Antila,Hanna Antila,Tomasz Róg,Mart Saarma,Clive R. Bramham,Claus Normann,Sari E. Lauri,Ilpo Vattulainen,Eero Castrén +26 more
TL;DR: In this paper, the transmembrane domain of TRKB, a BDNF receptor promoting neuronal plasticity and antidepressant responses, has a cholesterol-interaction site that may mediate the synaptic effects of cholesterol.

Small-molecule inhibitors of the RNA m6A demethylase FTO potently support the survival of dopamine neurons
Simona Selberg,Y. Li-Ying,Olesja Bondarenko,Esko Kankuri,Neinar Seli,Vera Kovaleva,Koit Herodes,Mart Saarma,Mati Karelson +8 more
TL;DR: In this article, a target-tailored development and biological characterization of small-molecule inhibitors of fat mass and obesity-associated protein (FTO), an RNA N6-methyladenosine (m6A) demethylase, is presented.
Posted ContentDOI
Antidepressants act by binding to the transmembrane domain of TRKB receptor
Plinio C. Casarotto,Mykhailo Girych,Senem Merve Fred,Rafael Moliner,Giray Enkavi,Caroline Biojone,Cecilia Cannarozzo,Cecilia A. Brunello,Frederike Winkel,Anna Steinzeig,Sudarshan Patil,Stefan Vestrig,Tsvetan Serchov,Vera Kovaleva,Cassiano R.A.F. Diniz,Liina Laukkanen,Iseline Cardon,Hanna Antila,Tomasz Róg,Mart Saarma,Clive R. Bramham,Claus Normann,Sari E. Lauri,Ilpo Vattulainen,Eero Castrén +24 more
TL;DR: It is suggested that binding to and facilitation of BDNF signaling through TRKB is the common mechanism for antidepressant action, which proposes a framework for how molecular effects of antidepressants are translated into clinical mood recovery.
