Suppression of MicroRNA-Silencing Pathway by HIV-1 During Virus Replication
Robinson Triboulet,Bernard Mari,Yea-Lih Lin,Christine Chable-Bessia,Yamina Bennasser,Kevin Lebrigand,Bruno Cardinaud,Thomas Maurin,Pascal Barbry,Vincent Baillat,Jacques Reynes,Pierre Corbeau,Kuan-Teh Jeang,Monsef Benkirane +13 more
TL;DR: It is shown that type III RNAses Dicer and Drosha, responsible for miRNA processing, inhibited virus replication both in peripheral blood mononuclear cells from HIV-1–infected donors and in latently infected cells.
Journal ArticleDOI
Suv39H1 and HP1γ are responsible for chromatin-mediated HIV-1 transcriptional silencing and post-integration latency
Isaure du Chéné,Euguenia Basyuk,Yea-Lih Lin,Robinson Triboulet,Anna Knezevich,Christine Chable-Bessia,Clément Mettling,Vincent Baillat,Jacques Reynes,Pierre Corbeau,Edouard Bertrand,Alessandro Marcello,Stéphane Emiliani,Rosemary Kiernan,Monsef Benkirane +14 more
TL;DR: It is shown that Suv39H1, HP1γ and histone H3Lys9 trimethylation play a major role in chromatin‐mediated repression of integrated HIV‐1 gene expression and that HIV‐ 1 reactivation could be achieved after HP 1γ RNA interference.
Journal ArticleDOI
SAMHD1 acts at stalled replication forks to prevent interferon induction
Flavie Coquel,Maria-Joao Silva,Maria-Joao Silva,Hervé Técher,Karina Zadorozhny,Sushma Sharma,Jadwiga Nieminuszczy,Clément Mettling,Elodie Dardillac,Antoine Barthe,Anne-Lyne Schmitz,Alexy Promonet,Alexandra Cribier,Amélie Sarrazin,Wojciech Niedzwiedz,Bernard S. Lopez,Vincenzo Costanzo,Lumir Krejci,Andrei Chabes,Monsef Benkirane,Yea-Lih Lin,Philippe Pasero +21 more
TL;DR: It is shown that SAMHD1 promotes degradation of nascent DNA at stalled replication forks in human cell lines by stimulating the exonuclease activity of MRE11, which activates the ATR–CHK1 checkpoint and allows the forks to restart replication.
Journal ArticleDOI
Senataxin resolves RNA:DNA hybrids forming at DNA double-strand breaks to prevent translocations.
Sarah Cohen,Nadine Puget,Yea-Lih Lin,Thomas Clouaire,Marion Aguirrebengoa,Vincent Rocher,Philippe Pasero,Yvan Canitrot,Gaëlle Legube +8 more
TL;DR: It is reported that senataxin is recruited at DNA double-strand breaks (DSBs) when they occur in transcriptionally active loci, and the helicase senatXin is identified to be involved in DNA repair and resolve RNA:DNA hybrids forming at DNAdouble-stranded breaks.
Journal ArticleDOI
SAMHD1 is mutated recurrently in chronic lymphocytic leukemia and is involved in response to DNA damage
Ruth Clifford,Tania Louis,Pauline Robbe,Sam Ackroyd,Adam Burns,Adele Timbs,Glen Wright Colopy,Helene Dreau,François Sigaux,Jean Gabriel Judde,Margalida Rotger,Amalio Telenti,Yea-Lih Lin,Philippe Pasero,Jonathan Maelfait,Michalis K. Titsias,Dena Cohen,Shirley Henderson,Mark T. Ross,David Bentley,Peter Hillmen,Andrew R. Pettitt,Jan Rehwinkel,Samantha J. L. Knight,Jenny C. Taylor,Yanick J. Crow,Monsef Benkirane,Anna Schuh +27 more
TL;DR: Evidence is provided that SAMHD1 regulates cell proliferation and survival and engages in specific protein interactions in response to DNA damage and that the presence of SAMHD 1 mutations in CLL promotes leukemia development.
