Journal ArticleDOI
Methylation of 12S rRNA is necessary for in vivo stability of the small subunit of the mammalian mitochondrial ribosome.
Metodi D. Metodiev,Nicole Lesko,Chan Bae Park,Yolanda Cámara,Yonghong Shi,Rolf Wibom,Kjell Hultenby,Claes M. Gustafsson,Nils-Göran Larsson +8 more
TL;DR: The results thus show that TFB1M is a nonredundant dimethyltransferase in mammalian mitochondria and provide a possible explanation for the universal conservation of adenine dimethylation of rRNA by showing a critical role in ribosome maintenance.
Journal ArticleDOI
LRPPRC is necessary for polyadenylation and coordination of translation of mitochondrial mRNAs
Benedetta Ruzzenente,Benedetta Ruzzenente,Metodi D. Metodiev,Anna Wredenberg,Ana Bratic,Ana Bratic,Chan Bae Park,Yolanda Cámara,Dusanka Milenkovic,Volker Zickermann,Rolf Wibom,Kjell Hultenby,Hediye Erdjument-Bromage,Paul Tempst,Ulrich Brandt,James B. Stewart,Claes M. Gustafsson,Nils-Göran Larsson,Nils-Göran Larsson +18 more
TL;DR: It is shown here that the leucine‐rich pentatricopeptide repeat containing (LRPPRC) gene is essential for embryonic development and point to the existence of an elaborate machinery that regulates mammalian mtDNA expression at the post‐transcriptional level.
Journal ArticleDOI
MTERF4 regulates translation by targeting the methyltransferase NSUN4 to the mammalian mitochondrial ribosome
Yolanda Cámara,Jorge Asin-Cayuela,Chan Bae Park,Metodi D. Metodiev,Yonghong Shi,Yonghong Shi,Benedetta Ruzzenente,Christian Kukat,Bianca Habermann,Rolf Wibom,Kjell Hultenby,Thomas Franz,Hediye Erdjument-Bromage,Paul Tempst,B. Martin Hallberg,Claes M. Gustafsson,Nils-Göran Larsson,Nils-Göran Larsson +17 more
TL;DR: The results thus show that MTERF4 is an important regulator of translation in mammalian mitochondria and directly controls mitochondrial ribosomal biogenesis and translation.
Journal ArticleDOI
MTERF3 Is a Negative Regulator of Mammalian mtDNA Transcription
Chan Bae Park,Jorge Asin-Cayuela,Yolanda Cámara,Yonghong Shi,Mina Pellegrini,Martina Gaspari,Rolf Wibom,Kjell Hultenby,Hediye Erdjument-Bromage,Paul Tempst,Maria Falkenberg,Claes M. Gustafsson,Nils-Göran Larsson +12 more
TL;DR: It is shown that MTERF3 is a negative regulator of mtDNA transcription initiation, the first example of a mitochondrial protein that acts as a specific repressor of mammalian mt DNA transcription initiation in vivo.
Journal ArticleDOI
Random Point Mutations with Major Effects on Protein-Coding Genes Are the Driving Force behind Premature Aging in mtDNA Mutator Mice
Daniel Edgar,Irina G. Shabalina,Yolanda Cámara,Anna Wredenberg,Maria Antonietta Calvaruso,Leo Nijtmans,Jan Nedergaard,Barbara Cannon,Nils-Göran Larsson,Aleksandra Trifunovic +9 more
TL;DR: It is reported that mitochondrial protein synthesis is unimpaired in mtDNA mutator mice consistent with the observed minor alterations of steady-state levels of mitochondrial transcripts, and these findings refute recent claims that circular mtDNA molecules with large deletions are driving the premature aging phenotype.
