Abstract: Nanoparticles (NP) are structures with at least one dimension of less than 100 nanometers (nm) and unique properties. Silver nanoparticles (AgNP), due to their bactericidal action, have found practical applications in medicine, cosmetics, textiles, electronics, and other fields. Nevertheless, their less advantageous properties which make AgNP potentially harmful to public health or the environment should also be taken into consideration. These nanoparticles are cyto- and genotoxic and accumulate in the environment, where their antibacterial properties may be disadvantageous for agriculture and waste management. The presented study reviews data concerning the biological effects of AgNP in mammalian cells in vitro: cellular uptake and excretion, localization in cellular compartments, cytotoxicity and genotoxicity. The mechanism of nanoparticle action consists on induction of the oxidative stress resulting in a further ROS generation, DNA damage and activation of signaling leading to various, cell type-specific pathways to inflammation, apoptotic or necrotic death. In order to assure a safe application of AgNP, further detailed studies are needed on the mechanisms of the action of AgNP on mammalian cells at the molecular level.

Abstract: Bacteriophage therapy is currently resurging as a potential complement/alternative to antibiotic treatment. However, preclinical evaluation lacks streamlined approaches. We here focus on preclinical approaches which have been implemented to assess bacteriophage efficacy against Pseudomonas biofilms and infections. Laser interferometry and profilometry were applied to measure biofilm matrix permeability and surface geometry changes, respectively. These biophysical approaches were combined with an advanced Airway Surface Liquid infection model, which mimics in vitro the normal and CF lung environments, and an in vivo Galleria larvae model. These assays have been implemented to analyze KTN4 (279,593 bp dsDNA genome), a type-IV pili dependent, giant phage resembling phiKZ. Upon contact, KTN4 immediately disrupts the P. aeruginosa PAO1 biofilm and reduces pyocyanin and siderophore production. The gentamicin exclusion assay on NuLi-1 and CuFi-1 cell lines revealed the decrease of extracellular bacterial load between 4 and 7 logs and successfully prevents wild-type Pseudomonas internalization into CF epithelial cells. These properties and the significant rescue of Galleria larvae indicate that giant KTN4 phage is a suitable candidate for in vivo phage therapy evaluation for lung infection applications.

Abstract: New results of the NA61/SHINE Collaboration at the CERN SPS on mean hadron multiplicities in proton-proton (p+p) interactions are analyzed within the transport models and the hadron resonance gas (HRG) statistical model. The chemical freeze-out parameters in p+p interactions and central Pb+Pb (or Au+Au) collisions are found and compared with each other in the range of the center-of-mass energy of the nucleon pair $\sqrt{{s}_{NN}}=3.2--17.3$ GeV. The canonical ensemble formulation of the HRG model is used to describe mean hadron multiplicities in p+p interactions and the grand canonical ensemble in central Pb+Pb and Au+Au collisions. The chemical freeze-out temperatures in p+p interactions are found to be larger than the corresponding temperatures in central nucleus-nucleus collisions.
