Abstract: The angiopoietins Ang-1 and Ang-2 have been identified as ligands of the receptor tyrosine kinase Tie-2 (refs. 1,2). Paracrine Ang-1-mediated activation of Tie-2 acts as a regulator of vessel maturation and vascular quiescence. In turn, the antagonistic ligand Ang-2 acts by an autocrine mechanism and is stored in endothelial Weibel-Palade bodies from where it can be rapidly released upon stimulation. The rapid release of Ang-2 implies functions of the angiopoietin-Tie system beyond its established role during vascular morphogenesis as a regulator of rapid vascular responses. Here we show that mice deficient in Ang-2 (encoded by the gene Angpt2) cannot elicit an inflammatory response in thioglycollate-induced or Staphylococcus aureus-induced peritonitis, or in the dorsal skinfold chamber model. Recombinant Ang-2 restores the inflammation defect in Angpt2(-/-) mice. Intravital microscopy showed normal TNF-alpha-induced leukocyte rolling in the vasculature of Angpt2(-/-)mice, but rolling cells did not firmly adhere to activated endothelium. Cellular experiments showed that Ang-2 promotes adhesion by sensitizing endothelial cells toward TNF-alpha and modulating TNF-alpha-induced expression of endothelial cell adhesion molecules. Together, these findings identify Ang-2 as an autocrine regulator of endothelial cell inflammatory responses. Ang-2 thereby acts as a switch of vascular responsiveness exerting a permissive role for the activities of proinflammatory cytokines.

Abstract: This article considers the problem of approximating a general asymptotically smooth function in two variables, typically arising in integral formulations of boundary value problems, by a sum of products of two functions in one variable. From these results an iterative algorithm for the low-rank approximation of blocks of large unstructured matrices generated by asymptotically smooth functions is developed. This algorithm uses only few entries from the original block and since it has a natural stopping criterion the approximative rank is not needed in advance.

Abstract: The primary structure of a voltage-dependent cal-cium channel from rabbit brain has been deduced by cloning and sequencing the complementary DNA. Calcium channel activity expressed from the cDNA is dramatically increased by coexpression of the α2 and β subunits, known to be associated with the dihydropyridine receptor. This channel is a high voltage-activated calcium channel that is insensitive both to nifedipine and to ω-conotoxin. We suggest that it is expressed predominantly in cerebellar Purkinje cells and granule cells.

Abstract: To find defects in software, one needs test cases that execute the software systematically, and oracles that assess the correctness of the observed behavior when running these test cases. This paper presents EvoSuite, a tool that automatically generates test cases with assertions for classes written in Java code. To achieve this, EvoSuite applies a novel hybrid approach that generates and optimizes whole test suites towards satisfying a coverage criterion. For the produced test suites, EvoSuite suggests possible oracles by adding small and effective sets of assertions that concisely summarize the current behavior; these assertions allow the developer to detect deviations from expected behavior, and to capture the current behavior in order to protect against future defects breaking this behavior.
