Abstract: Stimulating households to save energy with behaviour change support systems is a challenge and an opportunity to support efforts towards more sustainable energy consumption. The approaches developed so far, often either; do not consider the underlying behaviour change process in a systematic way, or do not provide a systematic linking of design elements to findings from behaviour change literature and the design of persuasive systems. This paper discusses the design and evaluation of a holistic socio-technical behaviour change system for energy saving that combines insights from behavioural theories and the persuasive system design in a systematic way. The findings from these two streams of research are combined into an integrated socio-technical model for informing the design of a behaviour change system for energy saving, which is then implemented in a concrete system design. The developed system combines smart meter data with interactive visualisations of energy consumption and energy saving impact, gamified incentive mechanisms, energy saving recommendations and attention triggers. The system design distinguishes between a version with non-personalized energy saving tips and a version with personalized recommendations that are deployed and evaluated separately. In this paper, we present the design and evaluation results of the non-personalized system in a real-world pilot. Obtained results indicate reduced energy consumption compared to a control group and a positive change in energy knowledge in the treatment group using the system, as well as positive user feedback about the suitability of the designed system to encourage energy saving.

Abstract: The purpose of this study was to establish whether participation in a teacher design team (TDT) is an effective way to increase the science teaching self-efficacy of primary school teachers who vary in their levels of experience and interest in science. A TDT is a group of at least 2 teachers from the same or related subjects working together to (re)design and enact (a part of) their common curriculum (A. Handelzalts, 2009). This study considers the context of a primary school in The Netherlands. Three forms of data were collected to obtain more in-depth data about the value of working in a TDT: the Dutch version of the Science Teaching Efficacy Belief Instrument (P. Fisser, B. Ormel, & C. Velthuis, 2010), an additional TDT questionnaire, and a focus group interview. The results indicate that professional development through a TDT can improve science teaching self-efficacy for teachers with varying levels of experience and interest

Abstract: Hormonally sensitive tissues, like the prostate, ovary, and breast, increasingly studied as targets of environmental chemicals, are sources of an enzyme potentially capable of transforming and activating xenobiotics to highly reactive metabolites. Our study specifically addresses the question of whether prostaglandin H synthase (PGHS) can activate phenolic metabolites of polychlorinated biphenyls (PCBs). We found that human recombinant PGHS-2 catalyzed the oxidation of ortho (2',3'- and 3',4'-) and para (2',5'-) dihydroxy 4-chlorobiphenyl metabolites to their corresponding quinones. These were trapped in situ with N-acetyl cysteine, and the reaction products were isolated and characterized by liquid chromatography coupled mass spectrometry and (1)H and heteronuclear ((1)H-(13)C) nuclear magnetic resonance spectroscopy. Both mono- and di-N-acetyl cysteine Michael addition adducts were identified, with the 2',3'- and 2',5'-dihydroxy metabolites predominantly forming mono-N-acetyl cysteine adducts, while the 3',4'-dihydroxy predominantly formed disubstituted N-acetyl cysteine adducts. These studies clearly demonstrate that the phenolic metabolites of these environmental pollutants are activated by PGHS, as cosubstrates, to highly reactive electrophilic PCB quinones, with a potential for protein and DNA damage, especially in nonhepatic tissues where the enzyme is found.

Abstract: Although a growing body of risk communication research focuses on how people process risk information, one question that is overlooked is how the seeking of information contributes to behavioral adaptation toward the risk issue. How are people’s behavioral responses to risks affected by the search for risk information? Building on the Framework of Risk Information Seeking (FRIS), this paper reports on two studies that focus on the experimental testing of several of the basic FRIS assumptions. In study 1, a 2 (involvement: high vs. low) × 2 (risk perception: high vs. low) between-subjects experiment was conducted to test the assumption that higher levels of involvement and risk perception stimulate the intention to seek additional risk information as well as the actual risk information. Study 2 is a partial replication of study 1. In study 2, a 2 (involvement: high vs. low) × 2 (fear appeal: present vs. absent) × 2 (response efficacy: high vs. low) between-subjects experiment was conducted to test how varying the levels of involvement, risk perception, and response efficacy influence actual and intended information seeking, as well as the intention to adopt risk-mitigating actions. The results showed that the high-involvement, high-risk perception, high-response efficacy group was most likely to actually seek information and make behavioral changes. The results are in accordance with basic FRIS assumptions. Implications for risk communication are discussed.

Abstract: DNA methylation plays a vital role in the cell, but loss-of-function mutations of the maintenance methyltransferase DNMT1 in normal human cells are lethal, precluding target identification, and existing hypomorphic lines are tumour cells. We generated instead a hypomorphic series in normal hTERT-immortalised fibroblasts using stably integrated short hairpin RNA. Approximately two-thirds of sites showed demethylation as expected, with one-third showing hypermethylation, and targets were shared between the three independently derived lines. Enrichment analysis indicated significant losses at promoters and gene bodies with four gene classes most affected: (1) protocadherins, which are key to neural cell identity; (2) genes involved in fat homoeostasis/body mass determination; (3) olfactory receptors and (4) cancer/testis antigen (CTA) genes. Overall effects on transcription were relatively small in these fibroblasts, but CTA genes showed robust derepression. Comparison with siRNA-treated cells indicated that shRNA lines show substantial remethylation over time. Regions showing persistent hypomethylation in the shRNA lines were associated with polycomb repression and were derepressed on addition of an EZH2 inhibitor. Persistent hypermethylation in shRNA lines was, in contrast, associated with poised promoters. We have assessed for the first time the effects of chronic depletion of DNMT1 in an untransformed, differentiated human cell type. Our results suggest polycomb marking blocks remethylation and indicate the sensitivity of key neural, adipose and cancer-associated genes to loss of maintenance methylation activity.
