Abstract: In an area endemic for visceral leishmaniasis, 16patients with human immunodeficiency virus (HIV) infection developed the disease. All belonged to populations at risk for AIDS (15 were intravenous drug abusers). Five patients fulfilled the criteria for full-blown AIDS, and two more fulfilled them after diagnosis of leishmaniasis. All presented with the classic manifestations of visceral leishmaniasis, but leishmania serology was negative in 15 patients (93%). Leishmania donovani amastigotes were identified in the bone marrow in all cases. Most patients responded initially to treatment with pentavalent antimonial drugs, but seven (43%) followed a chronic course, with multiple relapses in five, despite alternative treatments. Visceral leishmaniasis occurred in patients with different levels of depression of the CD4 to CD8 lymphocyte ratio. Mortality was 37% (six patients) and was independent of the chronic-relapsing course of the disease. In no case was leishmaniasis the primary cause of death. Our data establish that visceral leishmaniasis is an opportunistic infection in HIV-infected patients, and we suggest that in endemic areas it should be considered an indicator disease for the diagnosis of AIDS. © 1989 by The University of Chicago.

Abstract: The mechanism underlying diabetic osteopenia is still unclear and may involve osteoblastic activity and/or the deficit of insulin's anabolic action. Bone gla protein (BGP) is synthesized by the osteoblast and its synthesis increases with 1,25(OH)2D3 and fluoride. Because 1,25(OH)2D3 also stimulates insulin secretion, sodium fluoride administration can be used to investigate deficient osteoblastic activity in diabetics, as reflected by BGP levels. BGP was determined before and after administering sodium fluoride at a dosage of 50 mg/day/15 days to three groups: 14 patients with insulin-dependent diabetes, 16 diabetics on oral antidiabetic treatment, and 25 controls, all of similar age, sex, and characteristics. Basal BGP values (mean±SD) were low in diabetics on insulin treatment (4.3±1.1 ng/ml) and in diabetics on oral antidiabetics (5.8±1.2 ng/ml) as compared with controls (6.5±0.7 ng/ml) (P<0.001 and <0.05, respectively). After giving fluoride, BGP values did not change in the two diabetic groups but did vary in controls (8.1±0.6 ng/ml,P<0.001). These results suggest that deficient osteoblast function could be responsible for osteopenia in diabetics.

Abstract: Of 723 infertile men (128 with a history of cryptorchidism) whose testes were biopsied at the outer lateral face of the testis, five presented carcinoma in situ (CIS) in one testis. These testes were removed, serially sectioned, and examined by light microscopy. In order to evaluate whether only one or two biopsies are sufficient to diagnose CIS, before sectioning the testes four biopsies were taken at the anterior face, posterior face, superior pole, and inferior pole of the testis, respectively. Two of the five men had undergone orchiopexy in infancy and the testis contained tubules with Sertoli cells and isolated spermatogonia. CIS was also present in some tubules that were principally located near the rete testis. Of the four simulated biopsies, only that performed at the posterior face of the testis revealed CIS. The other three infertile men showed tubules with complete, although reduced, spermatogenesis, and tubules lined by Sertoli cells only. CIS was found in both types of tubules. These tubules with CIS formed lobules that extended throughout the testicular parenchyma. Most simulated biopsies performed in these three testes showed CIS. The average nuclear DNA content of CIS cells was about 4c in all testes. This content was similar both in tubules with complete spermatogenesis and in tubules with Sertoli cells only.

Abstract: The variations of the talar articular facets in 176 calcanei were studied and classified. Three types were considered: type A = calcanei with two articular facets for the talar head, with four subtypes; type B = calcanei with one articular facet for the talar head, and two subtypes, and type C = unique articular facies in the superior surface of the calcaneus for the talus. We found 53% (94 cases) type B calcanei and 46% (82 cases) type A calcanei. No calcanei of type C were seen.

Abstract: The interaction of some benzene and naphthalene derivatives with sodium dodecyl sulphate, hexadecyltrimethylmmonium bromide and polyoxyethylene [23] dodecanol micelles has been evaluated by high operformance liquid chromatography using micellar mobile phases. The micelle-solute association constants have been obtained for the compounds investigated. Good correlation between free energy of transfer for water-micelles and for octanol-water has been observed.

Abstract: The pattern of ethanol and acetaldehyde appearance in blood after an oral ethanol gavage (4 g/kg body wt) was not different at 12 or 21 days' gestation compared to virgin rats. Five min after maternal ethanol administration, concentrations of ethanol in fetal blood were lower than in maternal blood; however, at 15 min after ethanol administration, fetal and maternal blood levels were similar. Ethanol concentrations in fetal blood and amniotic fluid were already at equilibrium 5 min after ethanol administration. Acetaldehyde concentrations in fetal blood and in amniotic fluid were undetectable at all the times investigated, with the exception of fetuses from two pregnant rats studied 3 h after ethanol administration. Alcohol dehydrogenase activity in fetal liver and in placenta at late pregnancy was very low or undetectable, suggesting a very low rate of ethanol oxidation in vivo. After intravenous administration of acetaldehyde (10 mg/kg body wt), blood acetaldehyde concentrations were higher in pregnant than in virgin rats. Acetaldehyde concentrations in fetal blood and in amniotic fluid were similar to maternal blood at 2, 5, and 30 min after injection. When circulating concentrations of maternal and fetal acetaldehyde, obtained after either ethanol or acetaldehyde administration, were plotted, it was found that fetal blood concentrations of acetaldehyde were only detectable when maternal blood concentrations were greater than 80 microM. Concerning the acetaldehyde oxidation capacity, both the high and low affinity components of aldehyde dehydrogenase activity in fetal liver and placenta were very low as compared with maternal liver. However, aldehyde dehydrogenase activity in fetal liver was much higher than in placenta.(ABSTRACT TRUNCATED AT 250 WORDS)

Abstract: High 1,3-β-glucanase activity was detected during autolysis in a culture medium containing Penicillium oxalicum. It was due to the combined action of four enzymes. The purification process for the major enzyme produced a homogeneous band in the SDS polyacrylamide gel that corresponded to a molecular weight of 79 400 daltons. The enzyme pI was 6.3 and it was only active against 1,3-β-glucans, with a S0.5 of 0.23 mg ml−1 against laminarin. The enzymatic optima were found at pH 4 and 55°C, and instability was evident when pH and temperature were altered. The enzyme was not active against oxidated lamanarin and was barely inhibited by glucono-D-lactone. Hg2+, Ag+ and Fe2+ were effective inhibitors. The enzyme was adsorbed by concanavalin-A-sepharose.

Abstract: 1 In guinea-pig isolated taenia caeci and trachealis bathed in a K+-rich, Ca2+-free medium, CaCl2 (0.01-10 mM) produced a concentration-dependent contraction. Zn2+ (0.01-1 mM), Cd2+ (0.01-1 mM), verapamil (0.01-100 microM) and trifluoperazine (1-100 microM) were effective antagonists of CaCl2-induced responses. 2 Zn2+ and Cd2+ in concentrations from 0.01 to 1 mM were without effect on the tone of taenia and trachea in normal Tris solution. Conversely, Zn2+ and Cd2+, in concentrations of 1 mM, caused contraction of these tissues in a K+-rich, Ca2+-free medium. Zn2+ (1 mM)-induced contractions of taenia and trachea were completely inhibited by verapamil (10 microM). 3 In taenia and trachea skinned of their plasma membranes, tension development induced by Ca2+ (10 microM or 1 microM, respectively) was unaffected by verapamil (100 microM), whereas trifluoperazine (100 microM) depressed the maximal tension produced by Ca2+. Segments of skinned preparations contracted in response to low concentrations of Zn2+ (10 microM) or Cd2+ (10 microM). 4 It is concluded that Zn2+ may suppress Ca2+-induced spasm by a direct action on the binding sites of the Ca2+ channel.

Abstract: Studies were performed to determine whether and/or how dietary lipids participate in maternal hypertriglyceridemia during late gestation in the rat. After oral administration of glycerol-tri(1-14C)-palmitate, total radioactivity in plasma increased more rapidly in 20-day pregnant rats than in either 19-day pregnant rats or virgin controls. At the peak of plasma radioactivity, four hours after the tracer was administered, most of the plasma label corresponded to 14C-lipids in triglyceride-rich-lipoproteins (d < 1.006), and when expressed per micromol of triglyceride, values were higher in pregnant than in virgin rats. The difference was less after 24 hours, although at this time the level of 14C-lipids in d < 1.006 lipoproteins was still higher in 20-day pregnant rats than in virgins. Tissue 14C-lipids, as expressed per gram of fresh weight, were similar in pregnant and virgin rats, but the values in mammary glands were much higher in the former group. Estimated recovery of administered radioactivity four hours after tracer in total white adipose tissue, mammary glands, and plasma lipids was higher in pregnant than in virgin rats. No difference was found between 20-day pregnant and virgin rats either in the label retained in the gastrointestinal tract or in that exhaled as 14C-CO2 during the first four hours following oral administration of 14C-tripalmitate. These findings plus the known maternal hyperphagia, indicate that in the rat at late pregnancy triglyceride intestinal absorption is unchanged or even enhanced and that dietary lipids actively contribute to both maternal hypertriglyceridemia and lipid uptake by the mammary gland.

Abstract: Starch and polyacrylamide gel electrophoresis of seven isozyme systems was investigated as a means of identifying wild and cultivated species of Avena with different ploidy levels. By examining the characteristic isoenzymatic patterns, it was shown that there was considerable variability within the different species. However, it was nevertheless possible to unequivocally identify the species of wild oats and to distinguish between the different species belonging to the same genomic set, thus providing a definitive reference technique for the identification of Avena species in seed-testing laboratories. The relationships between Avena species were inferred from the electrophoresis data. The divergence of the A. maroccana - A. murphyi complex and its contribution to the AACC genomes are emphasized.

Abstract: An important topic in the neurobiology of the affective disorders relates to changes in the metabolism of neurotransmitters. Since the early 1960s, the ‘serotonergic’ hypothesis (Coppen et al, 1972) has been supported by findings such as a low concentration of 5-hydroxy-indoleacetic acid (5-HIAA) in the cerebro-spinal fluid (CSF) of patients with melancholic depression and/or suicidal behaviour (Banki & Arato, 1983; Asberg et al, 1984; Banki et al, 1984; Lopez-Ibor, Jr et al, 1985; Korpi et al, 1986) as well as the efficacy of serotonin precursors, when combined with MAOIs, in the treatment of depressed patients (Coppen et al, 1963; Glassman & Platman, 1969; Ayuso et al, 1971; Lopez-Ibor, Jr et al, 1976).

Abstract: Radiogrametry was done on 100 hands by two observers using two methods: (1) caliper and (2) magnifying glass measurement. At the mid-point of the second metacarpal, the total width (D) and medullary width (d) were measured and cortical thickness (C) was calculated from the difference (D-d). Statistical analysis disclosed that the intra-observer CV is smaller for both observers using the magnifying glass than with the caliper (1.9 and 2.1% vs. 2.8 and 4.3%, respectively). The inter-observer CV is also less with the magnifying glass (4.2 and 5.7% vs. 6.2 and 6.9%, respectively). Although measurements made with each method correlate significantly, the values (mean ± SD) obtained in isolated measurements using each method differ significantly, with C for both observers, as well as the intra-observer coefficient of variation (CV), being smaller. These results, which demonstrated different bone mass values as quantitated using a caliper or magnifying glass, indicate the greater reliability and precision of the latter method.

Abstract: The testes of 8 specimens of Triturus marmoratus were collected during each month of 1987 and processed for electron microscopy and light microscopy demonstration of testosterone (T) following the ABC (avidin-biotin peroxidase complex) method According to their staining affinity for anti-T antibodies, the glandular tissue cells were classified as T-, T+, T++, and T and the annual changes in the numbers of these cell populations, as well as in the volume occupied by the glandular tissue, were calculated The volume occupied by the glandular tissue increases from September to December; it begins to decrease in April and disappears from June to August The glandular tissue is formed from the interstitial cells that surround the lobules containing differentiating germ cells During the spermatogenic process, the interstitial cells do not show staining affinity for anti-T antibodies In August-September, the interstitial cells around the lobules that have completed spermatogenesis become positively stained (T+) and form the glandular tissue when the spermatozoa leave the testis The numbers of intensely stained cells in the glandular tissue (T++ and T ) increase from September to November; begin to decrease in December; disappear in January-February; increase again in March and decrease again in April until they disappear in June-September The interstitial cells, before their transformation in glandular tissue, are ultrastructurally similar to fibroblasts After their transformation these cells increase in size and develop abundant smooth endoplasmic reticulum, mitochondria with tubular cristae and lipid droplets This morphological pattern is maintained in the glandular tissue from September to April in spite of the changes in staining affinity during this period

Abstract: Adult male marbled newts (Triturus marmoratus) were collected at the beginning of the spermatogenetic period and exposed to different photoperiods (natural photoperiod with progressively increasing daylengths, total darkness, 8L:16D, 12L:12D, 16L:8D, and continuous light) for 3 months at 20°C. To evaluate the effect of photoperiodic input via pineal gland photoreceptors, two additional groups of newts were blinded by a non-aggressive method (an elastic rubber cap was adjusted to the head to cover the eyes but not the pineal photoreceptors). These animals were exposed either to the natural photoperiod or to 12 hr of light per day. Quantitative histologic studies on testicular development and germ-cell volume revealed no significant differences between non-blinded and blinded animals. Testicular size and germ-cell development increased in the following order: total darkness, constant light, 8L:12D, natural photoperiod, 12L:12D, and 16L:8D. These results suggest that (1) long photoperiods enhance testicular development, whereas short photoperiods or an environment of continuous light have the opposite effects and (2) the effect of photoperiods on testicular function in newts is independent of the ocular photoreceptors.
