TLDR
It is shown that caspase-3 is the primary executioner caspases in this system, necessary for cytochromec/dATP-inducible cleavage of fodrin, gelsolin, U1 small nuclear ribonucleoprotein, DNA fragmentation factor 45, and topoisomerase I, vimentin, Rb, and lamin B.

Apoptosis: A Review of Programmed Cell Death

TL;DR: The goal of this review is to provide a general overview of current knowledge on the process of apoptosis including morphology, biochemistry, the role of apoptoses in health and disease, detection methods, as well as a discussion of potential alternative forms of apoptotic proteins.

Mammalian caspases: structure ,a ctivation ,s ubstrates, and functions during apoptosis

TL;DR: Caspases, a family of cysteine-dependent aspartate-directed proteases, are prominent among the death proteases as discussed by the authors, and they play critical roles in initiation and execution of this process.

A decade of caspases

TL;DR: A large number of caspases from the cysteine proteases family play important roles in regulating apoptosis, and the mechanisms by which they mediate apoptosis and a variety of physiological and pathological processes are studied.

Caspase cleavage of tau: Linking amyloid and neurofibrillary tangles in Alzheimer's disease

TL;DR: A novel mechanism linking amyloid deposition and neurofibrillary tangles in AD is suggested: Aβ peptides promote pathological tau filament assembly in neurons by triggering caspase cleavage of tau and generating a proteolytic product with enhanced polymerization kinetics.
Journal ArticleDOI

Poly(ADP-ribose) polymerase-1 cleavage during apoptosis: an update.

TL;DR: During apoptosis, PARP-1 plays two opposite roles: its stimulation leads to poly(ADP-ribose) synthesis, whereas caspases cause PARP -1 cleavage and inactivation, and the fate and the possible role of these fragments during apoptosis will be discussed.

Apoptosis: a basic biological phenomenon with wide-ranging implications in tissue kinetics.

TL;DR: Apoptosis seems to be involved in cell turnover in many healthy adult tissues and is responsible for focal elimination of cells during normal embryonic development, and participates in at least some types of therapeutically induced tumour regression.

Cytochrome c and dATP-Dependent Formation of Apaf-1/Caspase-9 Complex Initiates an Apoptotic Protease Cascade

TL;DR: Mutation of the active site of caspase-9 attenuated the activation of cazase-3 and cellular apoptotic response in vivo, indicating that casp enzyme-9 is the most upstream member of the apoptotic protease cascade that is triggered by cytochrome c and dATP.

Caspases: Enemies Within

TL;DR: This work has shown that understanding caspase regulation is intimately linked to the ability to rationally manipulate apoptosis for therapeutic gain.

TL;DR: Apoptosis is a cell suicide mechanism that enables metazoans to control cell number in tissues and to eliminate individual cells that threaten the animal's survival.

Induction of apoptotic program in cell-free extracts : requirement for datp and cytochrome c

TL;DR: Cells undergoing apoptosis in vivo showed increased release of cy tochrome c to their cytosol, suggesting that mitochondria may function in apoptosis by releasing cytochrome c.
