Comparing the outcomes from three prospective multicenter studies investigating the use of CTC to monitor patients undergoing treatment for metastatic breast, colorectal, or prostate cancer is compared and the CTC definition used in these studies is reviewed.Abstract:
The increasing number of treatment options for patients with metastatic carcinomas has created a concomitant need for new methods to monitor their use. Ideally, these modalities would be noninvasive, be independent of treatment, and provide quantitative real-time analysis of tumor activity in a variety of carcinomas. Assessment of circulating tumor cells (CTCs) shed into the blood during metastasis may satisfy this need. We developed the CellSearch System to enumerate CTC from 7.5 mL of venous blood. In this review we compare the outcomes from three prospective multicenter studies investigating the use of CTC to monitor patients undergoing treatment for metastatic breast (MBC), colorectal (MCRC), or prostate cancer (MPC) and review the CTC definition used in these studies. Evaluation of CTC at anytime during the course of disease allows assessment of patient prognosis and is predictive of overall survival.read more

Cytokeratin 18 (CK18) and CK18 Fragments for Detection of Minimal Residual Disease in Colon Cancer Patients
TL;DR: There are no reliable methods to identify patients who are at increased risk of relapse and in need of adjuvant chemotherapy, and Appropriate biomarkers for the identification of CRC patients at high risk for recurrence and/or poor prognosis are required.

Novel method to detect, isolate, and culture prostate culturing circulating tumor cells
TL;DR: This study presents a fundamental advancement of CTC detection and culturing using a different mechanism (cell-cell interaction) rather than the traditional antibody-based immune-binding, such as CellSearchTM system, which has potential to be fully developed into a novel approach for early cancer metastasis detection, and chemotherapy efficacy monitoring.

Trapping and dynamic manipulation of magnetic contrast agent targeted cancer cells in photoacoustic imaging: Phantom study
TL;DR: It is demonstrated that targeted polystyrene beads can be successfully trapped in a recirculating system with flow rate of 12 ml/min and the manipulation of accumulated beads has been demonstrated to suppress undesirable background signals for sensitive and specific detection of targeted objects.

Circulating Tumor Cells in Breast Cancer: A Potential Liquid Biopsy
TL;DR: This chapter first introduces the principles of C TC isolation and detection methods, then the clinical utility of CTCs for prediction of prognosis and therapy response, and the heterogeneity ofCTCs is discussed.

The pathogenesis of cancer metastasis: the 'seed and soil' hypothesis revisited
TL;DR: It is now known that the potential of a tumour cell to metastasize depends on its interactions with the homeostatic factors that promote tumour-cell growth, survival, angiogenesis, invasion and metastasis.
