Journal ArticleDOI
Hepatic Mitochondrial Pyruvate Carrier 1 is Required for Efficient Regulation of Gluconeogenesis and Whole-body Glucose Homeostasis
Lawrence R. Gray,Mst Rasheda Sultana,Adam J. Rauckhorst,Lalita Oonthonpan,Sean C. Tompkins,Arpit Sharma,Xiaorong Fu,Ren Miao,Alvin D. Pewa,Kathryn S. Brown,Erin E. Lane,Ashley Dohlman,Diana Zepeda-Orozco,Jianxin Xie,Jared Rutter,Andrew W. Norris,James E. Cox,Shawn C. Burgess,Matthew J. Potthoff,Eric B. Taylor +19 more
TL;DR: It is demonstrated that the mitochondrial pyruvate carrier (MPC) comprising the Mpc1 and Mpc2 proteins is required for efficient regulation of hepatic gluc oneogenesis and that the MPC contributes to the elevated gluconeogenesis and hyperglycemia in T2D.
Journal ArticleDOI
Causal roles of mitochondrial dynamics in longevity and healthy aging.
TL;DR: It is proposed that altered mitochondrial dynamics might not just be a passive consequence of aging but might constitute an adaptive mechanism to mitigate age‐dependent cellular impairments and might be targeted to increase longevity and promote healthy aging.
Journal ArticleDOI
Disrupting Mitochondrial Pyruvate Uptake Directs Glutamine into the TCA Cycle away from Glutathione Synthesis and Impairs Hepatocellular Tumorigenesis.
Sean C. Tompkins,Ryan D. Sheldon,Adam J. Rauckhorst,Maria F. Noterman,Shane R. Solst,Jane L. Buchanan,Kranti A. Mapuskar,Alvin D. Pewa,Lawrence R. Gray,Lalita Oonthonpan,Arpit Sharma,Diego A Scerbo,Adam J. Dupuy,Douglas R. Spitz,Eric B. Taylor +14 more
TL;DR: The data show that liver MPC ablation markedly decreases tumorigenesis and that MPC-deficient tumors transcriptomically downregulate glutathione metabolism, raising the possibility that combining MPC disruption and glutATHione stress may be therapeutically useful in HCC and additional cancers.
Journal ArticleDOI
Metabolic Communication and Healthy Aging: Where Should We Focus Our Energy?
TL;DR: The pro-longevity effects of targeting metabolic pathways in specific tissues and how these interventions communicate with distal cells to modulate aging are discussed, which may be crucial in designing interventions that promote longevity without negative health consequences.
Journal ArticleDOI
Impaired skeletal muscle mitochondrial pyruvate uptake rewires glucose metabolism to drive whole-body leanness.
Arpit Sharma,Lalita Oonthonpan,Ryan D. Sheldon,Adam J. Rauckhorst,Zhiyong Zhu,Sean C. Tompkins,Kevin Cho,Wojciech J. Grzesik,Lawrence R. Gray,Diego A Scerbo,Alvin D. Pewa,Emily M. Cushing,Michael C. Dyle,James E. Cox,Christopher M. Adams,Brandon S.J. Davies,Richard K. Shields,Andrew W. Norris,Gary J. Patti,Leonid V. Zingman,Eric B. Taylor +20 more
TL;DR: The results illuminate the role of the skeletal muscle MPC as a whole-body carbon flux control point and highlight the potential utility of modulating muscle pyruvate utilization to ameliorate obesity and type 2 diabetes.
