Glutathione Primes T Cell Metabolism for Inflammation
Tak W. Mak,Tak W. Mak,Melanie Grusdat,Gordon S. Duncan,Catherine Dostert,Yannic Nonnenmacher,Yannic Nonnenmacher,Maureen A. Cox,Carole Binsfeld,Zhenyue Hao,Zhenyue Hao,Anne Brüstle,Momoe Itsumi,Christian Jäger,Ying Chen,Olaf Pinkenburg,Bärbel Camara,Markus Ollert,Carsten Bindslev-Jensen,Vasilis Vasiliou,Chiara Gorrini,Philipp A. Lang,Michael Lohoff,Isaac S. Harris,Karsten Hiller,Karsten Hiller,Dirk Brenner +26 more
TL;DR: It is reported that GSH is essential for T cell effector functions through its regulation of metabolic activity and plays an unexpected role in metabolic integration and reprogramming during inflammatory T cell responses.
Journal ArticleDOI
Glutathione Restricts Serine Metabolism to Preserve Regulatory T Cell Function.
Henry Kurniawan,Davide G. Franchina,Luana Guerra,Lynn Bonetti,Leticia Soriano Baguet,Melanie Grusdat,Lisa Schlicker,Oliver Hunewald,Catherine Dostert,Myriam P. Merz,Carole Binsfeld,Gordon S. Duncan,Sophie Farinelle,Yannic Nonnenmacher,Jillian Haight,Dennis Das Gupta,Anouk Ewen,Rabia Taskesen,Rashi Halder,Ying Chen,Christian Jäger,Markus Ollert,Paul Wilmes,Vasilis Vasiliou,Isaac S. Harris,Christiane B. Knobbe-Thomsen,Jonathan D. Turner,Tak W. Mak,Tak W. Mak,Tak W. Mak,Michael Lohoff,Johannes Meiser,Karsten Hiller,Dirk Brenner,Dirk Brenner +34 more
TL;DR: An unexpected role for GSH in restricting serine availability to preserve Treg functionality is revealed, using mice with Treg-specific ablation of the catalytic subunit of glutamate cysteine ligase.
Journal ArticleDOI
Glutathione-dependent redox balance characterizes the distinct metabolic properties of follicular and marginal zone B cells
Davide G. Franchina,Henry Kurniawan,Melanie Grusdat,Carole Binsfeld,Luana Guerra,Lynn Bonetti,Leticia Soriano-Baguet,Anouk Ewen,Takumi Kobayashi,Sophie Farinelle,Annamaria Minafra,Niels Vandamme,Anaïs Carpentier,Felix B. Kleine Borgmann,Christian Jäger,Ying Chen,Markus Kleinewietfeld,Vasilis Vasiliou,Michel Mittelbronn,Karsten Hiller,Philipp A. Lang,Dirk Brenner +21 more
TL;DR: In this paper , the authors showed that Gclc deficiency in the catalytic subunit of glutamate cysteine ligase (Gclc) is associated with increased ATP levels, glucose metabolism, mTOR activation, and protein synthesis.
Journal ArticleDOI
Protection From EAE in DOCK8 Mutant Mice Occurs Despite Increased Th17 Cell Frequencies in the Periphery
Alicia S. Wilson,Hsei Di Law,Christiane B. Knobbe-Thomsen,Conor J. Kearney,Jane Oliaro,Carole Binsfeld,Gaetan Burgio,Lora Starrs,Dirk Brenner,Katrina L. Randall,Anne Brüstle +10 more
TL;DR: This study shows that resistance to EAE in DOCK8 mutant mice is achieved despite a systemic Th17 bias, with Th17 cells remaining elevated systemically while cellular infiltration in the CNS was reduced.
Journal ArticleDOI
Pyruvate dehydrogenase fuels a critical citrate pool that is essential for Th17 cell effector functions.
Leticia Soriano-Baguet,Melanie Grusdat,Henry Kurniawan,Mohaned Benzarti,Carole Binsfeld,Anouk Ewen,Joseph Longworth,Lynn Bonetti,Luana Guerra,Davide G. Franchina,Takumi Kobayashi,Veronika Horkova,Charlène Verschueren,Sergio Helgueta,Deborah Gérard,Tushar H. More,Antonia Henne,Catherine Dostert,Sophie Farinelle,Antoine Lesur,Jean-Jacques Gérardy,Christian Jäger,Michel Mittelbronn,Lasse Sinkkonen,Karsten Hiller,Johannes Meiser,Dirk Brenner +26 more
TL;DR: In this paper , the importance of PDH function in T helper 17 (Th17) cells still remains to be studied, but it is shown that PDH is essential for the generation of a glucose-derived citrate pool needed for Th17 cell proliferation, survival, and effector function.
