IL‐6 up‐regulates Mcl‐1 in human myeloma cells through JAK / STAT rather than Ras / MAP kinase pathway
TL;DR: It is demonstrated that IL‐6, a survival factor for the human myeloma cell line MDN, rapidly up‐regulates Mcl‐1 whereas it has no effect on Bcl‐2 protein level and the involvement of the JAK / STAT pathway but not of the Ras’/’mitogen‐activated protein (MAP) kinase pathway in IL‐ 6‐induced M cl‐1 up‐regulation is suggested.
Journal ArticleDOI
Thrombin-induced endothelial microparticle generation: identification of a novel pathway involving ROCK-II activation by caspase-2.
Cédric Sapet,Stéphanie Simoncini,Béatrice Loriod,Denis Puthier,José Sampol,Catherine Nguyen,Françoise Dignat-George,Francine Anfosso +7 more
TL;DR: The involvement of caspase-2 in ROCK-II activation independently of cell death points out a novel signaling pathway that emphasizes the proteolytic activity of cospase in EMP generation in response to cell activation.
Journal ArticleDOI
Genome-wide characterization of mammalian promoters with distal enhancer functions
Lan T.M. Dao,Ariel O. Galindo-Albarrán,Jaime A. Castro-Mondragon,Charlotte Andrieu-Soler,Charlotte Andrieu-Soler,Charlotte Andrieu-Soler,Alejandra Medina-Rivera,Charbel Souaid,Guillaume Charbonnier,Aurélien Griffon,Laurent Vanhille,Tharshana Stephen,Tharshana Stephen,Jaafar Alomairi,David Martin,Magali Torres,Nicolas Fernandez,Eric Soler,Eric Soler,Eric Soler,Jacques van Helden,Denis Puthier,Salvatore Spicuglia +22 more
TL;DR: Extensive CRISPR–Cas9 genomic manipulation demonstrated the involvement of these promoters in the cis regulation of expression of distal genes in their natural loci and have important implications for the understanding of complex gene regulation in normal development and disease.
Journal ArticleDOI
Dynamic Competing Histone H4 K5K8 Acetylation and Butyrylation Are Hallmarks of Highly Active Gene Promoters
Afsaneh Goudarzi,Di Zhang,He Huang,Sophie Barral,Oh Kwang Kwon,Shankang Qi,Zhanyun Tang,Thierry Buchou,Anne-Laure Vitte,Tieming He,Zhongyi Cheng,Emilie Montellier,Jonathan Gaucher,Sandrine Curtet,Alexandra Debernardi,Guillaume Charbonnier,Denis Puthier,Carlo Petosa,Daniel Panne,Sophie Rousseaux,Robert G. Roeder,Yingming Zhao,Saadi Khochbin +22 more
TL;DR: This work investigates the butyrylation of histone H4 lysine 5 and 8 at gene promoters where acetylation guides the binding of Brdt, a bromodomain-containing protein, thereby mediating stage-specific gene expression programs and post-meiotic chromatin reorganization.
Journal ArticleDOI
High incidence of N and K-Ras activating mutations in multiple myeloma and primary plasma cell leukemia at diagnosis.
Stéphane Bézieau,Marie-Claire Devilder,Hervé Avet-Loiseau,Marie-Paule Mellerin,Denis Puthier,Erwan Pennarun,M. J. Rapp,Jean-Luc Harousseau,Jean-Paul Moisan,Régis Bataille +9 more
TL;DR: These early mutations could play a major role in the oncogenesis of MM and P‐PCL, and the validity of the technique was assessed by direct sequencing of cell lines and of some patients.
