Journal ArticleDOI

Sepp1UF forms are N-terminal selenoprotein P truncations that have peroxidase activity when coupled with thioredoxin reductase-1

TL;DR: A previously uncharacterized plasma form of Sepp1 is filtered in the glomerulus and taken up by renal proximal convoluted tubule (PCT) cells via megalin-mediated endocytosis, preventing loss of selenium in the urine and providing seenium for the synthesis of glutathione peroxidase-3.
Journal ArticleDOI

Inhibition of thioredoxin reductase 1 by porphyrins and other small molecules identified by a high-throughput screening assay.

TL;DR: A single-enzyme, dual-purpose assay for simultaneous identification of inhibitors and substrates of TrxR1 is described and rottlerin and the heme precursor protoporphyrin IX (PpIX) are found to be effective inhibitors of the enzyme.
Journal ArticleDOI

Details in the catalytic mechanism of mammalian thioredoxin reductase 1 revealed using point mutations and juglone-coupled enzyme activities.

TL;DR: It is found that a sole Cys residue at the C-terminal tail of TrxR1 is required for high-efficiency juglone-coupled NADPH oxidase activity of Sec-deficient enzyme, occurring with mixed one- and two-electron reactions producing superoxide.
Journal ArticleDOI

Rutin protects against H2O2-triggered impaired relaxation of placental arterioles and induces Nrf2-mediated adaptation in Human Umbilical Vein Endothelial Cells exposed to oxidative stress

TL;DR: Rutin functions as an antioxidant and is oxidized into a quinone that upregulates the Nrf2-mediated endogenous antioxidant response, which suggests that rutin selectively exerts its protective effects in regions with increased oxidative stress, and explains how r Rutin reduces the risk of developing CVD.
