TL;DR: Analyses of samples from patients with acute myeloid leukaemia reveal that drug response is associated with mutational status and gene expression; the generated dataset provides a basis for future clinical and functional studies of this disease.
Journal ArticleDOI
Molecularly targeted drug combinations demonstrate selective effectiveness for myeloid- and lymphoid-derived hematologic malignancies
Stephen E. Kurtz,Christopher A. Eide,Andy Kaempf,Vishesh Khanna,Samantha L. Savage,Angela Rofelty,Isabel English,Hibery Ho,Ravindra Nath Pandya,William J. Bolosky,Hoifung Poon,Michael W. Deininger,Robert H. Collins,Ronan T. Swords,Justin M. Watts,Daniel A. Pollyea,Bruno C. Medeiros,Elie Traer,Cristina E. Tognon,Motomi Mori,Brian J. Druker,Jeffrey W. Tyner +21 more
TL;DR: Ex vivo sensitivity profiling of patients with AML and CLL revealed specific patterns of ex vivo drug combination efficacy that were associated with select genetic, cytogenetic, and phenotypic disease subsets, highlighting the heuristic value of an integrated functional genomic approach to the identification of novel treatment strategies for hematologic malignancies.
Journal ArticleDOI
Targeting of colony-stimulating factor 1 receptor (CSF1R) in the CLL microenvironment yields antineoplastic activity in primary patient samples.

TL;DR: Results indicate that CSF1R small-molecule inhibitors target CD14-expressing monocytes in the CLL microenvironment, thereby depriving leukemia cells of extrinsic support signals and support the concept of simultaneously targeting supportive NLCs and CLL cells.
Journal ArticleDOI

Tyner +93 more
TL;DR: A large functional genomic resource is expanded, avenues for mechanistic exploration and drug development, and tools for predicting outcome in AML are revealed, including a single gene, PEAR1, which is found to be among the strongest predictors of patient survival.
Journal ArticleDOI
Colony Stimulating Factor 1 Receptor (CSF1R) As a Potential Novel Therapeutic Target in CLL
Tyler Sweeney,Stephen E. Spurgeon,Jeffrey W. Tyner,Anupriya Agarwal,Hibery Ho,Elie Traer,Patrice Lee,David Chantry,Brian J. Druker,Marc M. Loriaux +9 more
TL;DR: The results show that CSF1R is a potential therapeutic target in CLL and suggest that CLL supporting monocytes (nurse-like cells) express CSf1R and depend on CSF 1R for viability in a similar manner as TAMs depend onCSF1r in a variety of solid tumor settings.
