Journal ArticleDOI
A comparison of genetically matched cell lines reveals the equivalence of human iPSCs and ESCs.
Jiho Choi,Soohyun Lee,William Mallard,Kendell Clement,Guidantonio Malagoli Tagliazucchi,Guidantonio Malagoli Tagliazucchi,Hotae Lim,In Young Choi,Francesco Ferrari,Alexander M. Tsankov,Alexander M. Tsankov,Ramona Pop,Ramona Pop,Gabsang Lee,John L. Rinn,John L. Rinn,John L. Rinn,Alexander Meissner,Alexander Meissner,Peter J. Park,Konrad Hochedlinger,Konrad Hochedlinger +21 more
TL;DR: It is concluded that hESCs and hiPSCs are molecularly and functionally equivalent and cannot be distinguished by a consistent gene expression signature and genetic background variation is a major confounding factor for transcriptional and epigenetic comparisons of pluripotent cell lines.
Journal ArticleDOI
Generation of Multipotent Induced Neural Crest by Direct Reprogramming of Human Postnatal Fibroblasts with a Single Transcription Factor
Yong Jun Kim,Hotae Lim,Zhe Li,Yohan Oh,Irina Kovlyagina,In Young Choi,Xinzhong Dong,Xinzhong Dong,Gabsang Lee +8 more
TL;DR: This study reports direct reprogramming of human fibroblasts into induced neural crest (iNC) cells by overexpression of a single transcription factor, SOX10, in combination with environmental cues including WNT activation.
Journal ArticleDOI
Zika virus directly infects peripheral neurons and induces cell death.
Yohan Oh,Feiran Zhang,Yaqing Wang,Emily M. Lee,In Young Choi,Hotae Lim,Fahimeh Mirakhori,Ronghua Li,Luoxiu Huang,Tianlei Xu,Hao Wu,Cui Li,Cheng-Feng Qin,Zhexing Wen,Zhexing Wen,Qing-Feng Wu,Hengli Tang,Zhiheng Xu,Peng Jin,Hongjun Song,Hongjun Song,Guo Li Ming,Guo Li Ming,Gabsang Lee +23 more
TL;DR: It is shown that intraperitoneally or intraventricularly injected ZIKV in the mouse can infect and impact peripheral neurons in vivo, and ZikV productively infects stem-cell-derived human neural crest cells and peripheral neuron in vitro, leading to increased cell death, transcriptional dysregulation and cell-type-specific molecular pathology.
Journal ArticleDOI
Transcriptional landscape of myogenesis from human pluripotent stem cells reveals a key role of TWIST1 in maintenance of skeletal muscle progenitors
In Young Choi,In Young Choi,Hotae Lim,Hotae Lim,Hyeon Jin Cho,Yohan Oh,Bin Kuan Chou,Hao Bai,Linzhao Cheng,Yong Jun Kim,Sang-Hwan Hyun,Sang-Hwan Hyun,Hyesoo Kim,Joo Heon Shin,Gabsang Lee +14 more
TL;DR: This study characterized the transcriptional landscape of distinct human myogenic stages, and identified TWIST1 as a critical factor in maintenance of human PAX7::EGFP+ putative skeletal muscle progenitor cells.
Journal ArticleDOI
Comparison of three congruent patient-specific cell types for the modelling of a human genetic Schwann-cell disorder.
Bipasha Mukherjee-Clavin,Ruifa Mi,Barbara Kern,Barbara Kern,In Young Choi,Hotae Lim,Yohan Oh,Yohan Oh,B.M. Lannon,B.M. Lannon,Kevin J. Kim,Shaughn Bell,Junho K. Hur,Woochang Hwang,Young Hyun Che,Omer Habib,Robert H. Baloh,Kevin Eggan,Gerald Brandacher,Ahmet Hoke,Lorenz Studer,Yong Jun Kim,Gabsang Lee +22 more
TL;DR: A comparison of the molecular, cellular and functional characteristics of three congruent patient-specific cell types derived from currently available differentiation and direct-reprogramming technologies for the modelling of Charcot−Marie−Tooth 1A reveals commonly upregulated chemokines.
