TL;DR: Disclosed as discussed by the authors is a method for analyzing quantification of gene expression, which consists of purifying mRNA from total RNA to prepare fragmented mRNA; reverse transcription of the fragmented mRNA to prepare cDNA, purifying the cDNA to prepare blunt end DNA and then purising the blunt-end DNA fragments; and adding connector sequences to both ends of the DNA fragments with an "A" base added at an end thereof.
