Journal ArticleDOI
Mutations in PCSK9 cause autosomal dominant hypercholesterolemia.
Marianne Abifadel,Mathilde Varret,Jean-Pierre Rabès,Delphine Allard,Khadija Ouguerram,Martine Devillers,Corinne Cruaud,Suzanne Benjannet,Louise Wickham,D. Erlich,Aurélie Derré,Ludovic Villéger,Michel Farnier,Isabel Beucler,Eric Bruckert,Jean Chambaz,Bernard Chanu,Jean-Michel Lecerf,Gérald Luc,Philippe Moulin,Jean Weissenbach,Annick Prat,Michel Krempf,Claudine Junien,Nabil G. Seidah,Catherine Boileau +25 more
TL;DR: Two mutations in the gene PCSK9 (encoding proprotein convertase subtilisin/kexin type 9) that cause ADH are reported, a newly identified human subtilase that is highly expressed in the liver and contributes to cholesterol homeostasis.
Journal ArticleDOI
A mutation in the human leptin receptor gene causes obesity and pituitary dysfunction
Karine Clément,Christian Vaisse,Najiba Lahlou,Sylvie Cabrol,Véronique Pelloux,Dominique Cassuto,Micheline Gourmelen,Christian Dina,Jean Chambaz,Jean-Marc Lacorte,Arnaud Basdevant,Pierre Bougnères,Yves Lebouc,Philippe Froguel,Bernard Guy-Grand +14 more
TL;DR: A homozygous mutation in the human leptin receptor gene results in a truncated leptin receptor lacking both the transmembrane and the intracellular domains, which indicates that leptin is an important physiological regulator of several endocrine functions in humans.
Journal ArticleDOI
Resistance of colon cancer cells to long-term 5-fluorouracil exposure is correlated to the relative level of Bcl-2 and Bcl-X(L) in addition to Bax and p53 status.
Sabine Violette,Laurent Poulain,Elisabeth Dussaulx,Dominique Pepin,Anne-Marie Faussat,Jean Chambaz,Jean-Marc Lacorte,Cathy Staedel,Thécla Lesuffleur +8 more
TL;DR: The ability to resist to 5‐fluorouracil‐induced apoptosis of 8 colon cancer cell lines differing in p53 and bax status is reported and high levels of anti‐apoptotic Bcl‐2 and B cl‐xL proteins combined with a low level of Bax were correlated to high 5‐FU resistance of wild‐type p53 cell lines.
Journal ArticleDOI
Essential fatty acids interconversion in the human fetal liver.
TL;DR: The data suggest that arachidonic acid is supplied to the fetus through a preferential transfer across the placenta via delta 5- and delta 6-desaturase activities in fetal liver microsomes.
Journal ArticleDOI
Early Loss of E-cadherin from Cell-Cell Contacts Is Involved in the Onset of Anoikis in Enterocytes
Stéphane Fouquet,Veronica Haydée Lugo-Martinez,Anne-Marie Faussat,Flore Renaud,Philippe Cardot,Jean Chambaz,Martine Pinçon-Raymond,Sophie Thenet +7 more
TL;DR: The results stress the striking sensitivity of normal enterocytes to the loss of anchorage and the contribution of E-cadherin to the control of their survival/apoptosis balance and open new perspectives on the key role of this protein, which is dysregulated in the intestinal epithelium in both inflammatory bowel disease and cancer.
