Aaron Nilsen,Alexis N. LaCrue,Karen L. White,Isaac P. Forquer,R. Matthew Cross,Jutta Marfurt,Michael W. Mather,Michael J. Delves,David M. Shackleford,Fabián E. Sáenz,Joanne M. Morrisey,Jessica Anne Steuten,Tina Mutka,Yuexin Li,Grennady Wirjanata,Eileen Ryan,Sandra Duffy,Jane X. Kelly,Boni F. Sebayang,Anne-Marie Zeeman,Rintis Noviyanti,Robert E. Sinden,Clemens H. M. Kocken,Ric N. Price,Ric N. Price,Vicky M. Avery,Iñigo Angulo-Barturen,María Belén Jiménez-Díaz,Santiago Ferrer,Esperanza Herreros,Laura M. Sanz,Francisco-Javier Gamo,Ian Bathurst,Jeremy N. Burrows,Peter Siegl,R. Kiplin Guy,Rolf W. Winter,Akhil B. Vaidya,Susan A. Charman,Dennis E. Kyle,Roman Manetsch,Michael K. Riscoe +41 more
TL;DR: ELQ-300, a 4(1H)-quinolone-3-diarylether, which targets the liver and blood stages, including the forms that are crucial to disease transmission (gametocytes, zygotes, and ookinetes), has potential as a new drug for the treatment, prevention, and, ultimately, eradication of human malaria.
Journal ArticleDOI
Comparative antimalarial activities and ADME profiles of ozonides (1,2,4-trioxolanes) OZ277, OZ439, and their 1,2-dioxolane, 1,2,4-trioxane, and 1,2,4,5-tetraoxane isosteres.
Xiaofang Wang,Yuxiang Dong,Sergio Wittlin,Sergio Wittlin,Susan A. Charman,Francis C. K. Chiu,Jacques Chollet,Jacques Chollet,Kasiram Katneni,Janne Mannila,Julia Morizzi,Eileen Ryan,Christian Scheurer,Christian Scheurer,Jessica Anne Steuten,Josefina Santo Tomas,Josefina Santo Tomas,Christopher Snyder,Christopher Snyder,Jonathan L. Vennerstrom +19 more
TL;DR: In this article, the antimalarial activities and ADME profiles of the 1,2,4-trioxolane substructure of dispiro ozonides OZ277 and OZ439 were compared.
Journal ArticleDOI
Central nervous system exposure of next generation quinoline methanols is reduced relative to mefloquine after intravenous dosing in mice.
Geoffrey S. Dow,Erin Milner,Ian Bathurst,Jayendra B. Bhonsle,Diana Caridha,Sean Gardner,Lucia Gerena,Michael P. Kozar,Charlotte A. Lanteri,Anne Mannila,William McCalmont,Jay Moon,Kevin D. Read,Suzanne Norval,Norma Roncal,David M. Shackleford,Jason C. Sousa,Jessica Anne Steuten,Karen L. White,Qiang Zeng,Susan A. Charman +20 more
TL;DR: Reduction of CNS levels of NGQMs relative to mefloquine may be feasible and optimization of this property together with potency and long half-life may be practicable amongst diamine quinoline methanols.
Journal ArticleDOI
Brain uptake of diazepam and phenytoin in a genetic animal model of absence epilepsy.
Joseph A. Nicolazzo,Jessica Anne Steuten,Susan A. Charman,Nerida Taylor,Philip J. Davies,Steven Petrou,Steven Petrou +6 more
TL;DR: There is limited data available on changes to brain uptake of AEDs in spontaneous seizure animal models, such as genetic absence epilepsy, and changes in anti‐epileptic drugs used in these models are limited.
Comparative Antimalarial Activities and ADME Profiles of Ozonides
Yuxiang Dong,Sergio Wittlin,Susan A. Charman,Francis C. K. Chiu,Jacques Chollet,Kasiram Katneni,Janne Mannila,Julia Morizzi,Eileen Ryan,Christian Scheurer,Jessica Anne Steuten,Josefina Santo Tomas,Christopher Snyder,Jonathan L. Vennerstrom +13 more
TL;DR: The OZ439 series peroxides had superior exposure and efficacy in mice compared to the corresponding OZ277 series per oxides, and there was a good correlation between the antimalarial efficacy and ADME profiles in the rank order trioxolane > trioxane > tetraoxane.
