Journal ArticleDOI
Extensive germline genome engineering in pigs.
Yanan Yue,Weihong Xu,Yinan Kan,Hong-Ye Zhao,Yixuan Zhou,Xiaobin Song,Jiajia Wu,Juan Xiong,Dharmendra Goswami,Meng Yang,Lydia Lamriben,Mengyuan Xu,Qi Zhang,Yu Luo,Jianxiong Guo,Shengyi Mao,Deling Jiao,Tien Dat Nguyen,Zhuo Li,Jacob V. Layer,Mailin Li,Violette Paragas,Michele Youd,Zhongquan Sun,Yuan Ding,Weilin Wang,Hongwei Dou,Lingling Song,Xueqiong Wang,Lei Le,Xin Fang,Haydy George,Ranjith P. Anand,Shi Yun Wang,William F. Westlin,Marc Güell,James F. Markmann,Wenning Qin,Yangbin Gao,Hong-Jiang Wei,George M. Church,Luhan Yang +41 more
TL;DR: It is shown that pigs with all PERVs inactivated can also be genetically engineered to eliminate three xenoantigens and to express nine human transgenes that enhance the pigs' immunological compatibility and blood-coagulation compatibility with humans.
Journal ArticleDOI
Generation of pig induced pluripotent stem cells using an extended pluripotent stem cell culture system
Junjun Xu,Leqian Yu,Jianxiong Guo,Jinzhu Xiang,Zheng Zheng,Dengfeng Gao,Bingbo Shi,Haiyang Hao,Deling Jiao,Liang Zhong,Yu Wang,Jun Wu,Hong-Jiang Wei,Jianyong Han +13 more
TL;DR: These findings demonstrate that stable iPS cells could be generated in LCDM medium, which could give rise to both embryonic and extraembryonic cells in vivo, but the efficiency and level of chimeric contribution of pig LCDM-iPS cells were found low.
Journal ArticleDOI
Generation of biallelic knock-out sheep via gene-editing and somatic cell nuclear transfer.
Honghui Li,Gui Wang,Zhiqiang Hao,Guozhong Zhang,Yubo Qing,Shuanghui Liu,Lili Qing,Weirong Pan,Lei Chen,Guichun Liu,Ruoping Zhao,Baoyu Jia,Luyao Zeng,Jianxiong Guo,Lixiao Zhao,Heng Zhao,Chaoxiang Lv,Kaixiang Xu,Wenmin Cheng,Hushan Li,Hong-Ye Zhao,Wen Wang,Hong-Jiang Wei +22 more
TL;DR: These findings demonstrate the successful generation of MSTN biallelic-KO STH sheep via gene editing in somatic cells using TALEN technology and SCNT, and reveal that MSTn KO resulted in muscle fiber hypertrophy.
Journal ArticleDOI
Generation and characterization of stable pig pregastrulation epiblast stem cell lines.
Minglei Zhi,Jinying Zhang,Qianzi Tang,Dawei Yu,Shuai Gao,Dengfeng Gao,Pengliang Liu,Jianxiong Guo,Tang Hai,Jie Gao,Suying Cao,Zimo Zhao,Chongyang Li,Xiaogang Weng,Mengnan He,Tianzhi Chen,Yingjie Wang,Keren Long,Deling Jiao,Guanglei Li,Jiaman Zhang,Yan Liu,Yu Lin,Daxin Pang,Qianqian Zhu,Naixin Chen,Jingjing Huang,Xinze Chen,Yixuan Yao,Jingcang Yang,Zicong Xie,Xianya Huang,Mengxin Liu,Ran Zhang,Qiuyan Li,Yiliang Miao,Jianhui Tian,Xingxu Huang,Hongsheng Ouyang,Bofeng Liu,Wei Xie,Qi Zhou,Hong-Jiang Wei,Zhonghua Liu,Caihong Zheng,Mingzhou Li,Jianyong Han +46 more
TL;DR: In this article, a large-scale single-cell transcriptome analysis of pig embryos from embryonic day (E) 0 to E14, specifically, the tracing of pluripotency changes during epiblast development, was performed.
Journal ArticleDOI
Efficient generation of P53 biallelic knockout Diannan miniature pigs via TALENs and somatic cell nuclear transfer.
Shen Youfeng,Kaixiang Xu,Zaimei Yuan,Jianxiong Guo,Heng Zhao,Xuezeng Zhang,Lu Zhao,Yubo Qing,Honghui Li,Weirong Pan,Baoyu Jia,Hong-Ye Zhao,Hong-Jiang Wei +12 more
TL;DR: Although these genetically engineered Diannan miniature pigs had no tumorigenic signs, the P53 gene was dysfunctional, and it is believed that these pigs will provide powerful new resources for preclinical oncology and basic cancer research.
