Cancer exosomes perform cell-independent microRNA biogenesis and promote tumorigenesis.
Sonia A. Melo,Hikaru Sugimoto,Hikaru Sugimoto,Joyce T. O’Connell,Noritoshi Kato,Alberto Villanueva,August Vidal,Le Qiu,Edward Vitkin,Lev T. Perelman,Carlos A. Melo,Anthony Lucci,Cristina Ivan,George A. Calin,Raghu Kalluri,Raghu Kalluri +15 more
TL;DR: It is reported that breast cancer associated exosomes contain microRNAs (miRNAs) associated with the RISC-Loading Complex (RLC) and display cell-independent capacity to process precursor micro RNAs (pre-miRNas) into mature miRNAs.
Journal ArticleDOI
PGC-1α mediates mitochondrial biogenesis and oxidative phosphorylation in cancer cells to promote metastasis
Valerie S. LeBleu,Joyce T. O’Connell,Karina N. Gonzalez Herrera,Harriet Wikman,Klaus Pantel,Marcia C. Haigis,Fernanda Machado de Carvalho,Aline Santos Damascena,Ludmilla Thomé Domingos Chinen,Rafael Malagoli Rocha,John M. Asara,Raghu Kalluri +11 more
TL;DR: It is reported that migratory/invasive cancer cells specifically favour mitochondrial respiration and increased ATP production, and mitochondrial biogenesis and respiration induced by PGC-1α are also essential for functional motility of cancer cells and metastasis.
Journal ArticleDOI
Epigenetic Reprogramming of Cancer-Associated Fibroblasts Deregulates Glucose Metabolism and Facilitates Progression of Breast Cancer.
Lisa M. Becker,Joyce T. O’Connell,Annie P. Vo,Margo P. Cain,Désirée Tampe,Lauren Bizarro,Lauren Bizarro,Hikaru Sugimoto,Hikaru Sugimoto,Anna K. McGow,John M. Asara,John M. Asara,Sara Lovisa,Kathleen M. McAndrews,Rafal Zielinski,Philip L. Lorenzi,Michael Zeisberg,Sughra Raza,Valerie S. LeBleu,Valerie S. LeBleu,Valerie S. LeBleu,Raghu Kalluri +21 more
TL;DR: It is demonstrated that CAFs exhibit a metabolic shift toward lactate and pyruvate production and fuel biosynthetic pathways of cancer cells, and their breast cancer-promoting phenotype is partly mediated by oxygen-dependent epigenetic modifications.
Journal ArticleDOI
A Wnt-Bmp Feedback Circuit Controls Intertissue Signaling Dynamics in Tooth Organogenesis
Daniel J. O’Connell,Joshua W. K. Ho,Joshua W. K. Ho,Tadanori Mammoto,Annick Turbe-Doan,Joyce T. O’Connell,Psalm Haseley,Psalm Haseley,Samuel Koo,Samuel Koo,Nobuhiro Kamiya,Donald E. Ingber,Donald E. Ingber,Donald E. Ingber,Peter J. Park,Peter J. Park,Peter J. Park,Richard L. Maas +17 more
TL;DR: A systems biology approach that combined the generation and analysis of large-scale spatiotemporal gene expression data with mouse genetic experiments to gain insight into the mechanisms that control epithelial-mesenchymal signaling interactions in the developing mouse molar tooth identified a critical signaling circuit that controls the coordinated genome-wide expression changes and reciprocal signaling molecule dynamics.
