Kalfa,Shondra M. Pruett-Miller,Shengdar Q. Tsai,Mitchell J. Weiss,David R. Liu,David R. Liu,David R. Liu +47 more
TL;DR: This article used ABE8e-NRCH to convert the β-globin allele (HBBS) into Makassar βglobin (HBBG), a non-pathogenic variant.
Journal ArticleDOI
The autophagy-activating kinase ULK1 mediates clearance of free α-globin in β-thalassemia
Christophe Lechauve,Julia Keith,Eugene Khandros,Stephanie Fowler,Kalin Mayberry,Abdullah Freiwan,Christopher S. Thom,Paola Delbini,Paola Delbini,Emilio Boada Romero,Jingjing Zhang,Irene Motta,Irene Motta,Heather Tillman,M. Domenica Cappellini,M. Domenica Cappellini,Mondira Kundu,Mitchell J. Weiss +17 more
TL;DR: It is shown that loss of the autophagy-activating Unc-51–like kinase 1 (Ulk1) gene in β-thalassemic mice reduces autophagic clearance of α-globin in red blood cell precursors and exacerbates disease phenotypes, whereas inactivation of the canonical autophaky-related 5 (Atg5) gene has relatively minor effects.
Journal ArticleDOI
FBXO11-Mediated Proteolysis of BAHD1 Relieves PRC2-dependent Transcriptional Repression in Erythropoiesis.
Peng Xu,Daniel C. Scott,Beisi Xu,Yu Yao,Ruopeng Feng,Li Cheng,Kalin Mayberry,Yong-Dong Wang,Wenjian Bi,Lance E. Palmer,Moeko T. King,Hong Wang,Yuxin Li,Yiping Fan,Arno F. Alpi,Chunliang Li,Junmin Peng,James B Papizan,Shondra M. Pruett-Miller,Ria Spallek,Florian Bassermann,Yong Cheng,Brenda A. Schulman,Mitchell J. Weiss +23 more
TL;DR: It is shown that the E3 ubiquitin ligase FBXO11 relieves PRC2-mediated repression during erythroid maturation by targeting its newly identified substrate BAHD1, an H3K27me3 reader that recruits transcriptional co-repressors.
Journal ArticleDOI
Limitations of mouse models for sickle cell disease conferred by their human globin transgene configurations
Kaitly J. Woodard,Phillip A. Doerfler,Kalin Mayberry,Akshay Kumar Sharma,Rachel Levine,Jonathan Yen,Virginia Valentine,Lance E. Palmer,Marcus B. Valentine,Mitchell J. Weiss +9 more
TL;DR: The findings provide practical information on the genomic structures of two common mouse SCD models, illustrate their limitations for analyzing therapies to induce HbF and confirm the importance of distal DNA elements in human globin regulation.
Journal ArticleDOI
Adenosine Base Editing of γ-Globin Promoters Induces Fetal Hemoglobin and Inhibit Erythroid Sickling
Thiyagaraj Mayuranathan,Jonathan Yen,Gregory A. Newby,Gregory A. Newby,Gregory A. Newby,Yu Yao,Shaina N. Porter,Kaitly J. Woodard,Jingjing Zhang,Kalin Mayberry,Akshay Sharma,Shondra M. preutt-Miller,John F. Tisdale,Shengdar Q. Tsai,David R. Liu,David R. Liu,David R. Liu,Mitchell J. Weiss +17 more
TL;DR: Sharma et al. as discussed by the authors used ABE-based base editors to install precise nucleotide substitutions without double-stranded break (DSBs) in the tandem duplicated γ-globin genes, which can result in loss or inversion of the intervening genetic material and rearrangements.
