Targeting BCL-2 with the BH3 Mimetic ABT-199 in Estrogen Receptor-Positive Breast Cancer
François Vaillant,Delphine Merino,Delphine Merino,Lily Lee,Lily Lee,Kelsey Breslin,Bhupinder Pal,Bhupinder Pal,Matthew E. Ritchie,Matthew E. Ritchie,Gordon K. Smyth,Gordon K. Smyth,Michael Christie,Michael Christie,Michael Christie,Louisa J. Phillipson,Louisa J. Phillipson,Christopher J. Burns,Christopher J. Burns,G. Bruce Mann,G. Bruce Mann,Jane E. Visvader,Jane E. Visvader,Geoffrey J. Lindeman,Geoffrey J. Lindeman,Geoffrey J. Lindeman +25 more
TL;DR: Unexpectedly, BH3 mimetics were found to counteract the side effect of tamoxifen-induced endometrial hyperplasia and synergized with phosphatidylinositol 3-kinase (PI3K)/mammalian target of rapamycin (mTOR) inhibitors in eliciting apoptosis.
Journal ArticleDOI
Sensitization of BCL-2–expressing breast tumors to chemotherapy by the BH3 mimetic ABT-737
Samantha R. Oakes,François Vaillant,Elgene Lim,Lily Lee,Kelsey Breslin,Frank Feleppa,Siddhartha Deb,Matthew E. Ritchie,Elena A Takano,Teresa Ward,Stephen B. Fox,Daniele Generali,Gordon K. Smyth,Andreas Strasser,David C.S. Huang,Jane E. Visvader,Geoffrey J. Lindeman +16 more
TL;DR: In vivo evidence is provided that BH3 mimetics can be used to sensitize primary breast tumors to chemotherapy and further suggest that elevated BCL-2 expression constitutes a predictive response marker in breast cancer.
Journal ArticleDOI
Global changes in the mammary epigenome are induced by hormonal cues and coordinated by Ezh2.
Bhupinder Pal,Toula Bouras,Toula Bouras,Wei Shi,Wei Shi,François Vaillant,François Vaillant,Julie Sheridan,Julie Sheridan,Nai Yang Fu,Nai Yang Fu,Kelsey Breslin,Kun Jiang,Matthew E. Ritchie,Matthew E. Ritchie,Matthew D. Young,Geoffrey J. Lindeman,Geoffrey J. Lindeman,Geoffrey J. Lindeman,Gordon K. Smyth,Gordon K. Smyth,Jane E. Visvader,Jane E. Visvader +22 more
TL;DR: Findings reveal that Ezh2 couples hormonal stimuli to epigenetic changes that underpin progenitor activity, lineage specificity, and alveolar expansion in the mammary gland.
Journal ArticleDOI
Gata-3 Negatively Regulates the Tumor-Initiating Capacity of Mammary Luminal Progenitor Cells and Targets the Putative Tumor Suppressor Caspase-14
Marie Liesse Asselin-Labat,Kate D. Sutherland,François Vaillant,François Vaillant,David E. Gyorki,David E. Gyorki,Di Wu,Di Wu,Sheridan L Holroyd,Kelsey Breslin,Teresa Ward,Wei Shi,Wei Shi,Mary L. Bath,Siddhartha Deb,Stephen B. Fox,Gordon K. Smyth,Gordon K. Smyth,Geoffrey J. Lindeman,Geoffrey J. Lindeman,Geoffrey J. Lindeman,Jane E. Visvader,Jane E. Visvader +22 more
TL;DR: These studies identify GATA-3 as an important regulator of tumor initiation through its ability to promote the differentiation of committed luminal progenitor cells.
Journal ArticleDOI
Genome-wide binding and mechanistic analyses of Smchd1-mediated epigenetic regulation
Kelan Chen,Kelan Chen,Jiang Hu,Jiang Hu,Darcy Moore,Darcy Moore,Ruijie Liu,Sarah A. Kessans,Kelsey Breslin,Isabelle S Lucet,Isabelle S Lucet,Andrew Keniry,Andrew Keniry,Huei San Leong,Huei San Leong,Clare L. Parish,Douglas J. Hilton,Douglas J. Hilton,Richard J.L.F. Lemmers,Silvère M. van der Maarel,Peter E. Czabotar,Peter E. Czabotar,Renwick C. J. Dobson,Matthew E. Ritchie,Matthew E. Ritchie,Graham F. Kay,James M. Murphy,James M. Murphy,Marnie E. Blewitt,Marnie E. Blewitt +29 more
TL;DR: This study reveals, for the first time to the authors' knowledge, where Smchd1 binds genome-wide, its hitherto unappreciated functional interaction with chromatin organizer CCCTC-binding factor in gene regulation, and which part of the protein is required for chromatin binding, which leads to a new model of Smd1 function, where it directly binds DNA to mediate 3D chromatin architecture.
