Journal ArticleDOI
Association of γ-Secretase with Lipid Rafts in Post-Golgi and Endosome Membranes
Kulandaivelu S. Vetrivel,Haipeng Cheng,William Lin,Takashi Sakurai,Tong Li,Nobuyuki Nukina,Philip C. Wong,Huaxi Xu,Gopal Thinakaran +8 more
TL;DR: The association of all four components of the γ-secretase complex, namely presenilin 1 (PS1)-derived fragments, mature nicastrin, APH-1, and PEN-2, with cholesterol-rich detergent insoluble membrane (DIM) domains of non-neuronal cells and neurons that fulfill the criteria of lipid rafts is described.
Journal ArticleDOI
Membrane rafts in Alzheimer's disease beta-amyloid production.
TL;DR: The cell biology of APP, beta- and gamma-secretases and the data on their association with lipid rafts are summarized and potential raft targeting signals identified in the secretases and their importance on amyloidogenic processing of APP are discussed.
Journal ArticleDOI
Amyloidogenic processing of β-amyloid precursor protein in intracellular compartments
TL;DR: The salient features of Alzheimer disease amyloidogenesis are reviewed, the current knowledge on APP trafficking and isyloidogenic processing of APP in intracellular membrane compartments and microdomains is discussed, and β- and γ-secretases are discussed.
Journal ArticleDOI
Spatial segregation of γ-secretase and substrates in distinct membrane domains
Kulandaivelu S. Vetrivel,Haipeng Cheng,Seong-Hun Kim,Ying Chen,Natalie Y. Barnes,Ange‘le T. Parent,Sangram S. Sisodia,Gopal Thinakaran +7 more
TL;DR: It is suggested that the translocation of γ-secretase to lipid rafts in adults ensures processing of certain substrates, including APP CTFs, while limiting processing of other potential substrates.
Journal ArticleDOI
Axonal BACE1 dynamics and targeting in hippocampal neurons: a role for Rab11 GTPase
Virginie Buggia-Prevot,Celia G. Fernandez,Sean M. Riordan,Kulandaivelu S. Vetrivel,Jelita Roseman,Jack Waters,Vytautas P. Bindokas,Robert Vassar,Gopal Thinakaran +8 more
TL;DR: The results reveal novel information on dynamic BACE1 transport in neurons, and demonstrate that Rab11-GTPase function is critical for axonal sorting of Bace1.
