Benchtop isolation and characterization of functional exosomes by sequential filtration.
Mitja L. Heinemann,Matthias Ilmer,Leslie P. Silva,David H. Hawke,Alejandro Recio,Maria A. Vorontsova,Eckhard Alt,Jody Vykoukal +7 more
TL;DR: A straightforward, three-step protocol for exosome isolation of cell culture supernatants or large volumes of biofluid based on sequential steps of dead-end pre-filtration, tangential flow filtration (TFF), and low-pressure track-etched membrane filTration that is easy-to-implement and amenable to translation for clinical exosomes isolations.
Journal ArticleDOI
Measurement of DNA concentration as a normalization strategy for metabolomic data from adherent cell lines
Leslie P. Silva,Philip L. Lorenzi,Preeti Purwaha,Valeda Yong,David H. Hawke,John N. Weinstein +5 more
TL;DR: It is concluded that DNA concentration is a widely applicable method for normalizing metabolomic data from adherent cell lines.
Journal ArticleDOI
CRAGE enables rapid activation of biosynthetic gene clusters in undomesticated bacteria.
Gaoyan Wang,Zhiying Zhao,Jing Ke,Yvonne Engel,Yi-Ming Shi,David Robinson,Kerem Bingol,Zheyun Zhang,Benjamin P. Bowen,Benjamin P. Bowen,Katherine B. Louie,Bing Wang,Robert Evans,Yu Miyamoto,Kelly Cheng,Suzanne M. Kosina,Markus de Raad,Leslie P. Silva,Alicia Luhrs,Andrea Lubbe,David W. Hoyt,Charles Francavilla,Hiroshi Otani,Hiroshi Otani,Samuel Deutsch,Samuel Deutsch,Nancy M. Washton,Edward M. Rubin,Nigel J. Mouncey,Nigel J. Mouncey,Axel Visel,Axel Visel,Trent R. Northen,Trent R. Northen,Jan Fang Cheng,Jan Fang Cheng,Helge B. Bode,Yasuo Yoshikuni +37 more
TL;DR: Development of chassis-independent recombinase-assisted genome engineering (CRAGE) enables the integration of plasmids encoding biosynthetic gene clusters into the chromosomes of diverse bacteria to optimize production of natural products in non-native strains.
Journal ArticleDOI
An artifact in LC-MS/MS measurement of glutamine and glutamic acid: in-source cyclization to pyroglutamic acid.
TL;DR: The sensitivity and accuracy of Gln, Glu, and pGlu quantitation by electrospray ionization-based mass spectrometry can be improved dramatically by using chromatographic conditions that adequately separate the three metabolites, isotopic internal standards to correct for in-source pGLU formation, and user-optimized fragmentor voltage for acquisition of the MS spectra.
Journal ArticleDOI
Insight into the redox regulation of the phosphoglucan phosphatase SEX4 involved in starch degradation
Dylan M. Silver,Leslie P. Silva,Emmanuelle Issakidis-Bourguet,Mikkel A. Glaring,David C. Schriemer,Greg B. G. Moorhead +5 more
TL;DR: Results provide the first biochemical evidence for a redox‐dependent structural switch that regulates SEX4 activity, which represents the first plant phosphatase known to undergo reversible oxidation via disulfide bond formation like its mammalian counterparts.
