Journal ArticleDOI
TP53 and Decitabine in Acute Myeloid Leukemia and Myelodysplastic Syndromes
John S. Welch,Allegra A. Petti,Christopher A. Miller,Catrina Fronick,Michelle O'Laughlin,Robert S. Fulton,Richard K. Wilson,Jack Baty,Eric J. Duncavage,Bevan Tandon,Yi-Shan Lee,Lukas D. Wartman,Geoffrey L. Uy,Armin Ghobadi,Michael H. Tomasson,Iskra Pusic,Rizwan Romee,Todd A. Fehniger,Keith Stockerl-Goldstein,Ravi Vij,Stephen T. Oh,Camille N. Abboud,Amanda F. Cashen,Mark A. Schroeder,Meagan A. Jacoby,Sharon Heath,Kierstin Luber,Megan Janke,Andrew Hantel,Niloufer Khan,Madina Sukhanova,Randall W. Knoebel,Wendy Stock,Timothy A. Graubert,Matthew J. Walter,Peter Westervelt,Daniel C. Link,John F. DiPersio,Timothy J. Ley +38 more
TL;DR: Patients with AML and MDS who had cytogenetic abnormalities associated with unfavorable risk, TP53 mutations, or both had favorable clinical responses and robust (but incomplete) mutation clearance after receiving serial 10-day courses of decitabine.
Journal ArticleDOI
Clonal evolution underlying leukemia progression and Richter transformation in patients with ibrutinib-relapsed CLL.
Sabah Kadri,Jimmy Lee,Carrie Fitzpatrick,Natalie Galanina,Madina Sukhanova,Girish Venkataraman,Shruti Sharma,Brad Long,Kristin Petras,Megan Theissen,Mei Ming,Yuri Kobzev,Wenjun Kang,Ailin Guo,Weige Wang,Nifang Niu,Howard L. Weiner,Michael J. Thirman,Wendy Stock,Sonali M. Smith,Chadi Nabhan,Jeremy P. Segal,Pin Lu,Y. Lynn Wang +23 more
TL;DR: In patients who had Richter transformation, it is found that the transformed cells were clonal descendants of circulating leukemia cells but continued to undergo evolution and drifts, and transformed lymphoma cells in tissue may acquire a different BTK mutation from that in the CLL leukemia cells.
Journal ArticleDOI
Biogenic amines in Drosophila virilis under stress conditions.
TL;DR: In individuals of line 101 responding to stress by a hormonal stress reaction, the contents of DOPA, dopamine, octopamine, and Tyr were lower than those of line 147 that did not respond to the stress.
Journal ArticleDOI
Prognostic tumor sequencing panels frequently identify germ line variants associated with hereditary hematopoietic malignancies.
Michael W. Drazer,Sabah Kadri,Madina Sukhanova,Sushant A. Patil,Allison H. West,Simone Feurstein,Dalein A. Calderon,Matthew F. Jones,Caroline M. Weipert,Christopher K. Daugherty,Adrián A. Ceballos-López,Gordana Raca,Mark W. Lingen,Zejuan Li,Jeremy P. Segal,Jane E. Churpek,Lucy A. Godley +16 more
TL;DR: It is demonstrated that NGS-based prognostic panels may identify individuals at risk for HHMs despite not being designed for this purpose, and variants known to cause Li-Fraumeni syndrome as well as known pathogenic variants in genes such as DDX41 and GATA2 are especially likely to be of germ line origin.
Journal ArticleDOI
Activation of MYC, a bona fide client of HSP90, contributes to intrinsic ibrutinib resistance in mantle cell lymphoma
Jimmy Lee,Liang Leo Zhang,Wenjun Wu,Hui Guo,Yan Li,Madina Sukhanova,Girish Venkataraman,Shengjian Huang,Hui Zhang,Mir Alikhan,Pin Lu,Ailin Guo,Natalie Galanina,Jorge Andrade,Michael L. Wang,Y. Lynn Wang +15 more
TL;DR: It is demonstrated that MYC is a bona fide client of HSP90 in the context of MCL by both immunoprecipitation and chemical precipitation and can be inhibited via PU-H71 to overcome primary ibrutinib resistance.
