TL;DR: Treatment outcomes were significantly better with use of linezolid, later generation fluoroquinolones, bedaquiline, clofazimine, and carbapenems for treatment of multidrug-resistant tuberculosis, and the need for trials to ascertain the optimal combination and duration of these drugs is emphasised.
Journal ArticleDOI
The Peptidoglycan of Stationary-Phase Mycobacterium tuberculosis Predominantly Contains Cross-Links Generated by l,d-Transpeptidation
Marie Lavollay,Michel Arthur,Michel Arthur,Michel Arthur,Martine Fourgeaud,Martine Fourgeaud,Martine Fourgeaud,Lionel Dubost,Arul Marie,Nicolas Veziris,Didier Blanot,Laurent Gutmann,Jean-Luc Mainardi +12 more
TL;DR: It is shown that a complex adaptive response thought to endow M. tuberculosis with the capacity to survive several months of combinatorial antibiotic treatment may involve remodeling of the peptidoglycan network by substitution of 4-->3 cross-links generated by the D,D-transpeptidase activity of penicillin-binding proteins by 3-->3Cross- links generated by a transpePTidase of L,D specificity.
Journal ArticleDOI
Assessment of Clarithromycin Susceptibility in Strains Belonging to the Mycobacterium abscessus Group by erm(41) and rrl Sequencing
Sylvaine Bastian,Nicolas Veziris,Anne-Laure Roux,Florence Brossier,Jean-Louis Gaillard,Vincent Jarlier,Emmanuelle Cambau +6 more
TL;DR: Clarithromycin-resistant strains, including the six rrl mutants, were more often isolated in cystic fibrosis patients, but this was not significantly associated with a previous treatment.
Journal ArticleDOI
Novel Gyrase Mutations in Quinolone-Resistant and -Hypersusceptible Clinical Isolates of Mycobacterium tuberculosis: Functional Analysis of Mutant Enzymes
Alexandra Aubry,Alexandra Aubry,Nicolas Veziris,Emmanuelle Cambau,Chantal Truffot-Pernot,Vincent Jarlier,L. Mark Fisher +6 more
TL;DR: Describing several multidrug-resistant clinical isolates of M. tuberculosis carrying mutations in the genes encoding the GyrA or GyrB subunits associated with quinolone resistance or hypersusceptibility found that several isolates bearing a novel combination of gyrA T80A and A90G changes were hypersuceptible to ofloxacin.
Journal ArticleDOI
Detection by GenoType MTBDRsl Test of Complex Mechanisms of Resistance to Second-Line Drugs and Ethambutol in Multidrug-Resistant Mycobacterium tuberculosis Complex Isolates
Florence Brossier,Nicolas Veziris,Alexandra Aubry,Vincent Jarlier,Wladimir Sougakoff,Wladimir Sougakoff +5 more
TL;DR: The GenoType MTBDRsl test efficiently detects the most common mutations involved in resistance to fluoroquinolones, aminoglycosides/cyclic peptide, and ethambutol and accurately assesses susceptibility to amikacin.
