Journal ArticleDOI
A genome-wide map of diversity in Plasmodium falciparum.
Sarah K. Volkman,Pardis C. Sabeti,David DeCaprio,Daniel E. Neafsey,Stephen F. Schaffner,Danny A. Milner,Johanna P. Daily,Ousmane Sarr,Daouda Ndiaye,Omar Ndir,S. Mboup,Manoj T. Duraisingh,Amanda K. Lukens,Alan Derr,Nicole Stange-Thomann,Skye G. Waggoner,Robert C. Onofrio,Liuda Ziaugra,Evan Mauceli,Sante Gnerre,David B. Jaffe,Joanne Zainoun,Roger C. Wiegand,Bruce W. Birren,Daniel L. Hartl,James E. Galagan,Eric S. Lander,Dyann F. Wirth,Dyann F. Wirth +28 more
TL;DR: An initial survey of genetic variation across the P. falciparum genome is described and its potential utility in identifying genes subject to recent natural selection and in understanding the population genetics of this parasite is demonstrated.
Journal ArticleDOI
Distinct physiological states of Plasmodium falciparum in malaria-infected patients
Johanna P. Daily,Daniel Scanfeld,Nathalie Pochet,Nathalie Pochet,K. G. Le Roch,David Plouffe,Michael Kamal,Ousmane Sarr,S. Mboup,Omar Ndir,David Wypij,K. Levasseur,Edwin L. Thomas,Pablo Tamayo,Carolyn K. Dong,Yingyao Zhou,Eric S. Lander,Daouda Ndiaye,Dyann F. Wirth,Elizabeth A. Winzeler,Elizabeth A. Winzeler,Jill P. Mesirov,Aviv Regev +22 more
TL;DR: A large study of in vivo expression profiles of parasites derived directly from blood samples from infected patients reveals a previously unknown physiological diversity in the in vivo biology of the malaria parasite, and indicates in vivo and in vitro studies to determine how this variation may affect disease manifestations and treatment.
Journal ArticleDOI
A Systematic Map of Genetic Variation in Plasmodium falciparum
Claire Kidgell,Sarah K. Volkman,Johanna P. Daily,Justin O. Borevitz,David Plouffe,Yingyao Zhou,Jeffrey R. Johnson,Karine G. Le Roch,Ousmane Sarr,Omar Ndir,S. Mboup,Serge Batalov,Dyann F. Wirth,Elizabeth A. Winzeler,Elizabeth A. Winzeler +14 more
TL;DR: It is speculated that an amplification of the GTP cyclohydrolase enzyme in the folate biosynthesis pathway may increase flux through this pathway and facilitate parasite resistance to antifolate drugs.
Journal ArticleDOI
Genome-wide SNP genotyping highlights the role of natural selection in Plasmodium falciparum population divergence
Daniel E. Neafsey,Stephen F. Schaffner,Sarak K. Volkman,Sarak K. Volkman,Daniel J. Park,Philip Montgomery,Danny A. Milner,Amanda K. Lukens,David I. Rosen,Rachel F. Daniels,Nathan Houde,Joseph F. Cortese,Erin Tyndall,Casey Gates,Nicole Stange-Thomann,Ousmane Sarr,Daouda Ndiaye,Omar Ndir,S. Mboup,Marcelo U. Ferreira,Sandra do Lago Moraes,Aditya P Dash,Chetan E. Chitnis,Roger C. Wiegand,Daniel L. Hartl,Bruce W. Birren,Eric S. Lander,Pardis C. Sabeti,Dyann F. Wirth +28 more
TL;DR: The distinct population profile of nonsynonymous variants indicates that natural selection has a significant influence on genomic diversity in P. falciparum, and that many of these changes may reflect functional variants deserving of follow-up study.
Journal ArticleDOI
A molecular epidemiological study of var gene diversity to characterize the reservoir of Plasmodium falciparum in humans in Africa
Donald Chen,Alyssa E. Barry,Aleksandra Leliwa-Sytek,Terry Ann Smith,Ingrid Peterson,Stuart M. Brown,Florence Migot-Nabias,Philippe Deloron,Moses Kortok,Kevin Marsh,Johanna P. Daily,Johanna P. Daily,Daouda Ndiaye,Ousmane Sarr,Souleymane Mboup,Karen P. Day,Karen P. Day +16 more
TL;DR: The findings show that var typing as a molecular surveillance system defines the extent of genetic complexity in the reservoir of infection to complement measures of malaria prevalence and suggest that var genetics could greatly inform current malaria mapping approaches and predict complex malaria population dynamics.
