Journal ArticleDOI
Prediction and Reduction of the Aggregation of Monoclonal Antibodies
Rob van der Kant,Anne R. Karow-Zwick,Joost Van Durme,Michaela Blech,Rodrigo Gallardo,Daniel Seeliger,Kerstin Aßfalg,Pieter Baatsen,Griet Compernolle,Ann Gils,Joey Studts,Patrick Schulz,Patrick Garidel,Joost Schymkowitz,Joost Schymkowitz,Frederic Rousseau,Frederic Rousseau +16 more
TL;DR: This work rationally design variants that display a marked decrease in aggregation propensity while retaining antigen binding through the introduction of artificial aggregation gatekeeper residues, showing that reducing protein aggregation is beneficial throughout the development process.
Journal ArticleDOI
Hsp90 Mediates Membrane Deformation and Exosome Release.
Elsa Lauwers,Elsa Lauwers,Yu-Chun Wang,Yu-Chun Wang,Rodrigo Gallardo,Rob van der Kant,Emiel Michiels,Jef Swerts,Jef Swerts,Pieter Baatsen,Pieter Baatsen,Samantha S. Zaiter,Shelli R. McAlpine,Natalia V. Gounko,Natalia V. Gounko,Frederic Rousseau,Joost Schymkowitz,Patrik Verstreken,Patrik Verstreken +18 more
TL;DR: This work structurally separated the Hsp90 membrane-deforming function from its well-characterized chaperone activity, and shows that this previously unrecognized function is required for exosome release.
Journal ArticleDOI
Structural hot spots for the solubility of globular proteins
Ashok Ganesan,Ashok Ganesan,Aleksandra Siekierska,Aleksandra Siekierska,Jacinte Beerten,Jacinte Beerten,Jacinte Beerten,Marijke Brams,Joost Van Durme,Joost Van Durme,Greet De Baets,Greet De Baets,Rob van der Kant,Rob van der Kant,Rodrigo Gallardo,Rodrigo Gallardo,Meine Ramakers,Meine Ramakers,Tobias Langenberg,Tobias Langenberg,Hannah Wilkinson,Hannah Wilkinson,Frederik De Smet,Frederik De Smet,Chris Ulens,Frederic Rousseau,Frederic Rousseau,Joost Schymkowitz,Joost Schymkowitz +28 more
TL;DR: It is shown that mutations at specific positions within a protein structure can act as APR suppressors without affecting protein stability, suggesting that mutational suppression of APRs provides a simple strategy to increase protein solubility.
Journal ArticleDOI
Post-translational Control of the Temporal Dynamics of Transcription Factor Activity Regulates Neurogenesis
Xiao-Jiang Quan,Liqun Yuan,Luca Tiberi,Annelies Claeys,Natalie De Geest,Jiekun Yan,Rob van der Kant,Rob van der Kant,Wei R. Xie,Wei R. Xie,Tiemo J. Klisch,Tiemo J. Klisch,Joost Shymkowitz,Joost Shymkowitz,Frederic Rousseau,Frederic Rousseau,Mathieu Bollen,Monique Beullens,Huda Y. Zoghbi,Pierre Vanderhaeghen,Pierre Vanderhaeghen,Bassem A. Hassan +21 more
TL;DR: Using genome editing in Drosophila, it is shown that Atonal outlives its mRNA but is inactivated by phosphorylation, which causes quantitative changes in expression dynamics and target gene expression resulting in neuronal number and fate defects.
Journal ArticleDOI
Solubis: a webserver to reduce protein aggregation through mutation.
Joost Van Durme,Greet De Baets,Rob van der Kant,Meine Ramakers,Ashok Ganesan,Hannah Wilkinson,Rodrigo Gallardo,Frederic Rousseau,Frederic Rousseau,Joost Schymkowitz,Joost Schymkowitz +10 more
TL;DR: This work provides access to an automated pipeline to identify mutations that reduce protein aggregation by reducing the intrinsic aggregation propensity of the sequence using the TANGO algorithm, while taking care not to disrupt the thermodynamic stability of the native structure (using the empirical force-field FoldX).
