Journal ArticleDOI
Tregs prevent GVHD and promote immune reconstitution in HLA-haploidentical transplantation
Mauro Di Ianni,Mauro Di Ianni,Franca Falzetti,Alessandra Carotti,Adelmo Terenzi,Flora Castellino,Elisabetta Bonifacio,Beatrice Del Papa,Tiziana Zei,Roberta Iacucci Ostini,Debora Cecchini,Teresa Aloisi,Katia Perruccio,Loredana Ruggeri,Chiara Balucani,Antonio Pierini,Paolo Sportoletti,Cynthia Aristei,Brunangelo Falini,Yair Reisner,Andrea Velardi,Franco Aversa,Massimo F. Martelli +22 more
TL;DR: It is shown for the first time in humans that adoptive transfer of Tregs prevented GVHD in the absence of any posttransplantation immunosuppression, promoted lymphoid reconstitution, improved immunity to opportunistic pathogens, and did not weaken the graft-versus-leukemia effect.
Journal ArticleDOI
HLA-haploidentical transplantation with regulatory and conventional T-cell adoptive immunotherapy prevents acute leukemia relapse
Massimo F. Martelli,Mauro Di Ianni,Mauro Di Ianni,Loredana Ruggeri,Franca Falzetti,Alessandra Carotti,Adelmo Terenzi,Antonio Pierini,Maria Speranza Massei,Lucia Amico,Elena Urbani,Beatrice Del Papa,Tiziana Zei,Roberta Iacucci Ostini,Debora Cecchini,Rita Tognellini,Yair Reisner,Franco Aversa,Brunangelo Falini,Andrea Velardi +19 more
TL;DR: It is demonstrated the immunosuppressive potential of Tregs can be used to suppress GVHD without loss of the benefits of graft-versus-leukemia (GVL) activity, and humanized murine models provided insights into the mechanisms underlying separation of GVL from GV HD.
Journal ArticleDOI
T regulatory cell separation for clinical application
Mauro Di Ianni,Mauro Di Ianni,Beatrice Del Papa,Tiziana Zei,Roberta Iacucci Ostini,Debora Cecchini,Maria Grazia Cantelmi,Stefano Baldoni,Paolo Sportoletti,Laura Cavalli,Alessandra Carotti,Antonio Pierini,Brunangelo Falini,Massimo F. Martelli,Franca Falzetti +14 more
TL;DR: Cells isolated by means of this approach can be used in allogeneic hematopoietic stem cell transplantation to reduce the incidence and severity of GvHD without bystander inhibition of general immunity.
Journal ArticleDOI
Clinical-Grade–Expanded Regulatory T Cells Prevent Graft-versus-Host Disease While Allowing a Powerful T Cell–Dependent Graft-versus-Leukemia Effect in Murine Models
Beatrice Del Papa,Loredana Ruggeri,Elena Urbani,Stefano Baldoni,Debora Cecchini,Tiziana Zei,Roberta Iacucci Ostini,Barbara Crescenzi,Alessandra Carotti,Antonio Pierini,Paolo Sportoletti,Paolo Bartolomeo,Franca Falzetti,Cristina Mecucci,Andrea Velardi,Massimo F. Martelli,Mauro Di Ianni +16 more
TL;DR: A good manufacturing practices-compatible expansion protocol to improve number and purity of regulatory T cells (Tregs) available for clinical trials is developed and might represent the basis for treating high-risk leukemia and/or relapsed/refractory leukemia patients with high-dose Treg/Tcons.
Journal ArticleDOI
Graft engineering for allogeneic haploidentical stem cell transplantation.
Antonio Tabilio,Franca Falzetti,Tiziana Zei,Maria De Ioanni,Elisabetta Bonifacio,Feliciana Battelli,Roberta Iacucci Ostini,Stelvio Ballanti,Michele Cimminiello,Monia Capponi,Carla Silvani,Olivia Minelli,Katia Fettucciari,Pierfrancesco Marconi,Emanuela Rosati,Antonella Santucci,Mauro Di Ianni,Franco Aversa,Massimo F. Martelli +18 more
TL;DR: Progress in graft processing has ensured primary, sustained engraftment rates of over 90% and has significantly reduced the incidence of severe acute GvHD and EBV-related lymphoproliferative disorders.
