Aneuploidy renders cancer cells vulnerable to mitotic checkpoint inhibition
Yael Cohen-Sharir,James M. McFarland,Mai Abdusamad,Carolyn Marquis,Sara Vanessa Bernhard,Mariya Kazachkova,Helen Tang,Marica Rosaria Ippolito,Kathrin Laue,Johanna Zerbib,Heidi L.H. Malaby,Andrew Jones,Lisa-Marie Stautmeister,Irena Bockaj,René Wardenaar,Nicholas J. Lyons,Ankur K. Nagaraja,Ankur K. Nagaraja,Adam J. Bass,Adam J. Bass,Diana C.J. Spierings,Floris Foijer,Rameen Beroukhim,Rameen Beroukhim,Stefano Santaguida,Stefano Santaguida,Todd R. Golub,Todd R. Golub,Jason Stumpff,Zuzana Storchova,Uri Ben-David +30 more
TL;DR: In this paper, aneuploid cancer cells exhibited aberrant spindle geometry and dynamics, and kept dividing when the SAC was inhibited, resulting in the accumulation of mitotic defects, and in unstable and less-fit karyotypes.
Journal ArticleDOI
Genetic instability from a single S phase after whole-genome duplication
Simon Gemble,René Wardenaar,Kristina Keuper,Nishit Srivastava,Maddalena Nano,Anne-Sophie Macé,Andréa E. Tijhuis,Sara Vanessa Bernhard,Diana C.J. Spierings,Anthony Simon,Oumou Goundiam,Helfrid Hochegger,Matthieu Piel,Floris Foijer,Zuzana Storchova,Renata Basto +15 more
TL;DR: In this paper , the authors used DNA combing and single-cell sequencing to show that DNA replication dynamics is perturbed, generating under-and over-replicated regions in the first S phase following tetraploidy.
Journal ArticleDOI
Modelling chromosome structural and copy number changes to understand cancer genomes.
TL;DR: The obtained findings reveal that genomic alterations strongly affect the cellular physiology and, importantly, instigate further genomic instability.
Posted ContentDOI
Whole-genome duplication shapes the aneuploidy landscape of human cancers
Prasad K,Mathew Bloomfield,Hagai Levi,Kristina Keuper,Sara Vanessa Bernhard,Nicolaas C. Baudoin,Leor G,Maybelline Giam,Cheng Kit Wong,Giulia Rancati,Zuzana Storchova,Daniela Cimini,Uri Ben-David +12 more
TL;DR: In this article, the authors analyzed clinical tumor samples from 449 WGD- tumors and 157 WGD+ tumors across 22 tumor types and found distinct recurrence patterns of aneuploidy in both WGD and non-WGD+ tumor types.
Posted ContentDOI
Selective vulnerability of aneuploid human cancer cells to inhibition of the spindle assembly checkpoint
Yael Cohen-Sharir,James M. McFarland,Mai Abdusamad,Carolyn Marquis,Helen Tang,Marica Rosaria Ippolito,Sara Vanessa Bernhard,Kathrin Laue,Heidi L.H. Malaby,Andrew Jones,Mariya Kazachkova,Nicholas J. Lyons,Ankur K. Nagaraja,Ankur K. Nagaraja,Adam J. Bass,Adam J. Bass,Rameen Beroukhim,Rameen Beroukhim,Stefano Santaguida,Stefano Santaguida,Jason Stumpff,Todd R. Golub,Todd R. Golub,Zuzana Storchova,Uri Ben-David +24 more
TL;DR: A novel synthetic lethal interaction between aneuploidy and the SAC, which may have direct therapeutic relevance for the clinical application of SAC inhibitors is revealed and its cellular and molecular underpinnings are explored.
