Journal ArticleDOI
Copper induces cell death by targeting lipoylated TCA cycle proteins
Peter Tsvetkov,Shannon Coy,Boryana Petrova,Margaret Dreishpoon,A. Verma,Mai Abdusamad,Jordan Rossen,Lena M. Joesch-Cohen,Ranad Humeidi,Ryan Spangler,John K. Eaton,Evgeni M. Frenkel,Mustafa Koçak,Steven M. Corsello,Svetlana Lutsenko,Naama Kanarek,Sandro Santagata,Todd R. Golub +17 more
TL;DR: It is shown in human cells that copper-dependent, regulated cell death is distinct from known death mechanisms and is dependent on mitochondrial respiration, and that excess copper promotes the aggregation of lipoylated proteins and links mitochondrial metabolism to copper- dependent death.
Journal ArticleDOI
Cas9 activates the p53 pathway and selects for p53-inactivating mutations.
Oana M. Enache,Veronica Rendo,Mai Abdusamad,Daniel D. Lam,Desiree Davison,Sangita Pal,Naomi Currimjee,Julian M. Hess,Sasha Pantel,Anwesha Nag,Aaron R. Thorner,John G. Doench,Francisca Vazquez,Rameen Beroukhim,Rameen Beroukhim,Todd R. Golub,Uri Ben-David,Uri Ben-David +17 more
TL;DR: Genetic and transcriptional consequences of Cas9 expression induces DNA damage and activates the p53 pathway, and it can lead to the selection of cells with p53-inactivating mutations, and Cas9 is less active in wild-type TP53 cell lines than in TP53- mutant cell lines.
Journal ArticleDOI
Aneuploidy renders cancer cells vulnerable to mitotic checkpoint inhibition
Yael Cohen-Sharir,James M. McFarland,Mai Abdusamad,Carolyn Marquis,Sara Vanessa Bernhard,Mariya Kazachkova,Helen Tang,Marica Rosaria Ippolito,Kathrin Laue,Johanna Zerbib,Heidi L.H. Malaby,Andrew Jones,Lisa-Marie Stautmeister,Irena Bockaj,René Wardenaar,Nicholas J. Lyons,Ankur K. Nagaraja,Ankur K. Nagaraja,Adam J. Bass,Adam J. Bass,Diana C.J. Spierings,Floris Foijer,Rameen Beroukhim,Rameen Beroukhim,Stefano Santaguida,Stefano Santaguida,Todd R. Golub,Todd R. Golub,Jason Stumpff,Zuzana Storchova,Uri Ben-David +30 more
TL;DR: In this paper, aneuploid cancer cells exhibited aberrant spindle geometry and dynamics, and kept dividing when the SAC was inhibited, resulting in the accumulation of mitotic defects, and in unstable and less-fit karyotypes.
Journal ArticleDOI
Synthetic Lethal Interaction between the ESCRT Paralog Enzymes VPS4A and VPS4B in Cancers Harboring Loss of Chromosome 18q or 16q.

Golub,Francisca Vazquez,Francisca Vazquez,Andrew J. Aguirre,Andrew J. Aguirre +49 more
TL;DR: A compendium of synthetic lethal vulnerabilities is described and V PS4A and VPS4B are nominated as high-priority therapeutic targets for cancers with 18q or 16q loss.

Phosphate dysregulation via the XPR1:KIDINS220 protein complex is a therapeutic vulnerability in ovarian cancer
Daniel P. Bondeson,Brenton R. Paolella,Adhana Asfaw,Michael V. Rothberg,Thomas A. Skipper,Carly Langan,Alfredo Gonzalez,Lauren E. Surface,Kentaro Ito,Mariya Kazachkova,William Colgan,Allison Warren,Josh Dempster,Mike Burger,Maria Ericsson,Andrew Tang,Iris Fung,Emily Chambers,Mai Abdusamad,Nancy Dumont,John G. Doench,Federica Piccioni,David E. Root,Jesse S. Boehm,William C. Hahn,William C. Hahn,Michael Mannstadt,James M. McFarland,Francisca Vazquez,Todd R. Golub,Todd R. Golub +30 more
TL;DR: All data point to the XPR1:KIDINS220 complex - and phosphate dysregulation more broadly - as a therapeutic vulnerability in ovarian cancer.
