Identification and characterization of a novel human histone H3 lysine 36-specific methyltransferase
Xiao-Jian Sun,Xiao-Jian Sun,Ju Wei,Xinyan Wu,Ming Hu,Lan Wang,Lan Wang,Hai Hong Wang,Qinghua Zhang,Sai Juan Chen,Sai Juan Chen,Qiu Hua Huang,Zhu Chen,Zhu Chen +13 more
TL;DR: The results suggest that HYPB HMTase may coordinate histone methylation and transcriptional regulation in mammals and open perspective for the further study of the potential roles of HYPB protein in hematopoiesis and pathogenesis of HD.
Journal ArticleDOI
The leukemogenicity of AML1-ETO is dependent on site-specific lysine acetylation.
Lan Wang,Alexander Gural,Xiao-Jian Sun,Xinyang Zhao,Fabiana Perna,Gang Huang,Megan A. Hatlen,Ly P. Vu,Fan Liu,Haiming Xu,Takashi Asai,Hao Xu,Tony DeBlasio,Silvia Menendez,Francesca Voza,Yanwen Jiang,Philip A. Cole,Jinsong Zhang,Ari Melnick,Robert G. Roeder,Stephen D. Nimer +20 more
TL;DR: This work has found that AML1-ETO, the fusion protein generated by the t(8;21) translocation, is acetylated by the transcriptional coactivator p300 in leukemia cells isolated from t( 8; 21) AML patients, and that this acetylation is essential for its self-renewal–promoting effects in human cord blood CD34+ cells and its leukemogenicity in mouse models.
Journal ArticleDOI
SET1 and p300 Act Synergistically, through Coupled Histone Modifications, in Transcriptional Activation by p53
Zhanyun Tang,Wei Yi Chen,Miho Shimada,Uyen T. T. Nguyen,Jaehoon Kim,Xiao-Jian Sun,Toru Sengoku,Robert K. McGinty,Joseph Fernandez,Tom W. Muir,Tom W. Muir,Robert G. Roeder +11 more
TL;DR: A mechanism in which SET1C and p300 act cooperatively, through direct interactions and coupled histone modifications, to facilitate the function of p53 is established.
Journal ArticleDOI
Hematopoietic gene expression profile in zebrafish kidney marrow
Huai-Dong Song,Xiao-Jian Sun,Xiao-Jian Sun,Min Deng,Guo-Wei Zhang,Yi Zhou,Xinyan Wu,Yan Sheng,Yi Chen,Zheng Ruan,Chun-Lei Jiang,Hui-Yong Fan,Leonard I. Zon,John P. Kanki,Ting Xi Liu,A. Thomas Look,Zhu Chen +16 more
TL;DR: The results indicate that conserved genetic programs regulate vertebrate hematopoiesis and vasculogenesis, and support the role of the zebrafish as an important animal model for studying both normal development and the molecular pathogenesis of human blood diseases.
Journal ArticleDOI
A stable transcription factor complex nucleated by oligomeric AML1–ETO controls leukaemogenesis
Xiao-Jian Sun,Zhanxin Wang,Lan Wang,Lan Wang,Yanwen Jiang,Nils Kost,David Soong,Wei Yi Chen,Zhanyun Tang,Tomoyoshi Nakadai,Olivier Elemento,Wolfgang Fischle,Ari Melnick,Dinshaw J. Patel,Stephen D. Nimer,Stephen D. Nimer,Robert G. Roeder +16 more
TL;DR: New mechanisms of action of AML1–ETO are revealed, and a potential therapeutic target in t(8;21)-positive acute myeloid leukaemia is provided.
