Variations in GABRA2, Encoding the α2 Subunit of the GABAA Receptor, Are Associated with Alcohol Dependence and with Brain Oscillations
Howard J. Edenberg,Danielle M. Dick,Xiaoling Xuei,Huijun Tian,Laura Almasy,Lance O. Bauer,Raymond R. Crowe,Alison Goate,Victor Hesselbrock,Kevin A. Jones,Jennifer M. Kwon,Ting-Kai Li,John I. Nurnberger,Sean O'Connor,Theodore Reich,John P. Rice,Marc A. Schuckit,Bernice Porjesz,Tatiana Foroud,Henri Begleiter +19 more
TL;DR: The very strong association of GABRA2 with both alcohol dependence and the beta frequency of the electroencephalogram, combined with biological evidence for a role of this gene in both phenotypes, suggest that GABra2 might influence susceptibility to alcohol dependence by modulating the level of neural excitation.
Journal ArticleDOI
Variants in nicotinic receptors and risk for nicotine dependence
Laura J. Bierut,Jerry A. Stitzel,Jen C. Wang,Anthony L. Hinrichs,Richard A. Grucza,Xiaoling Xuei,Nancy L. Saccone,Scott F. Saccone,Sarah Bertelsen,Louis Fox,William J. Horton,Naomi Breslau,John P. Budde,C. Robert Cloninger,Danielle M. Dick,Tatiana Foroud,Dorothy K. Hatsukami,Victor Hesselbrock,Eric O. Johnson,John Kramer,Samuel Kuperman,Pamela A. F. Madden,Kevin Mayo,John I. Nurnberger,Ovide F. Pomerleau,Bernice Porjesz,Oliver Reyes,Marc A. Schuckit,Gary E. Swan,Jay A. Tischfield,Howard J. Edenberg,John P. Rice,Alison Goate +32 more
TL;DR: This study confirms that at least two independent variants in this nicotinic receptor gene cluster contribute to the development of habitual smoking in some populations, and it underscores the importance of multiple genetic variants contributing to theDevelopment of common diseases in various populations.
Journal ArticleDOI
Genome-wide association study of alcohol dependence implicates a region on chromosome 11
Howard J. Edenberg,Daniel L. Koller,Xiaoling Xuei,Leah Wetherill,Jeanette N. McClintick,Laura Almasy,Laura J. Bierut,Kathleen K. Bucholz,Alison Goate,Fazil Aliev,Danielle M. Dick,Victor Hesselbrock,Anthony L. Hinrichs,John Kramer,Samuel Kuperman,John I. Nurnberger,John P. Rice,Marc A. Schuckit,Robert E. Taylor,B. Todd Webb,Jay A. Tischfield,Bernice Porjesz,Tatiana Foroud +22 more
TL;DR: Combining evidence from the case-control study, the follow-up in families, and gene expression provided strongest support for the association of a cluster of genes on chromosome 11 with alcohol dependence.
Journal ArticleDOI
Association of alcohol dehydrogenase genes with alcohol dependence: a comprehensive analysis
Howard J. Edenberg,Xiaoling Xuei,Hui Ju Chen,Huijun Tian,Leah Wetherill,Danielle M. Dick,Laura Almasy,Laura J. Bierut,Kathleen K. Bucholz,Alison Goate,Victor Hesselbrock,Samuel Kuperman,John I. Nurnberger,Bernice Porjesz,John P. Rice,Marc A. Schuckit,Jay A. Tischfield,Henri Begleiter,Tatiana Foroud +18 more
TL;DR: There was strong evidence that variations in ADH4 are associated with alcoholism: 12 SNPs were significantly associated and the region of strongest association ran from intron 1 to 19.5 kb beyond the 3' end of the gene.
Journal ArticleDOI
The Role of GABRA2 in Risk for Conduct Disorder and Alcohol and Drug Dependence across Developmental Stages
Danielle M. Dick,Laura J. Bierut,Anthony L. Hinrichs,Louis Fox,Kathleen K. Bucholz,John Kramer,Samuel Kuperman,Victor Hesselbrock,Marc A. Schuckit,Laura Almasy,Jay A. Tischfield,Bernice Porjesz,Henri Begleiter,John I. Nurnberger,Xiaoling Xuei,Howard J. Edenberg,Tatiana Foroud +16 more
TL;DR: In a sample of children and adolescents ascertained as part of the COGA project, it is found that GABRA2 is significantly associated with childhood conduct disorder symptoms, but not with childhood alcohol dependence symptoms.
