TL;DR: It is shown that Treg cell development was achieved by the combination of two independent processes, i.e., the expression of Foxp3 and the establishment of TReg cell-specific CpG hypomethylation pattern, and those T cells in which the two events have concurrently occurred are developmentally set into the T Reg cell lineage.

TREG-cell therapies for autoimmune rheumatic diseases.
TL;DR: Three potential strategies for the clinical use of TREG cells to treat autoimmune rheumatic disease are discussed: expansion of self-antigen-specific natural Treg cells in vivo; propagation of antigen- specific natural TREG Cells ex vivo, by in vitro antigenic stimulation, and subsequent transfer back into the host.
