SIRT6 rescues the age related decline in base excision repair in a PARP1-dependent manner.

TL;DR: The mechanistic study suggests that, similar to the regulatory role that SIRT6 plays in DNA DSB repair, SIRT 6 regulates BER in a PARP1-depdendent manner, and overexpression of Sirt6 rescues the decline of BER with age in aged fibroblasts.

TL;DR: Rescue experiments indicate that restoration of XRCC4 and Lig4 may suppress the onset of stress-induced premature cellular senescence, suggesting that improving NHEJ efficiency and fidelity by targeting the N HEJ pathway holds great potential to delay aging and mitigate aging-related pathologies.

A PARP1-BRG1-SIRT1 Axis Promotes HR Repair by Reducing Nucleosome Density at DNA Damage Sites

TL;DR: It is demonstrated that PARP1 is required for HR by modulating nucleosome density at damage sites, and it is reported that the efficiency of HR but not NHEJ negatively correlates with nucleosomes density.
