Abstract: Serum estrone, estradiol-1 7!) and progesterone were measured in bitches in which LH, sexual behavior and laparoscopically confirmed time of ovulation had been previously studied. Steroid values from 25 estrous periods were normalized to day of peak LH (Day 0). Estrone, estradiol-17(3 and progesterone concentration did not differ significantly in either dogs subjected to laparoscopy at 48 h intervals or those bitches undergoing no laparoscopy. During the preovulatory interval, mean peripheral estrone concentrations were more variable than estradiol-1713; however, both estrogens tended to rise gradually throughout this time period. Overall maximal estradiol-1 713 concentration was detected on Day -2 with estrone peaking later (Day -0.5). Both estrogens declined (P<0.05) coincidentally with the LH surge. Progesterone from Days -14.0 through -1.5 was maintained at basal concentrations. An increase (P<0.05) in mean progesterone occurred on the day of the preovulatory LI-I peak which correlated with direct observation of apparent preovulatory follicle luteinization. Progesterone increased gradually from Day -0.5 through 8 and then varied for the remainder of the sampling period. Analysis of individual cycles indicated that circulating serum concentrations of reproductive steroids fluctuated from day to day or within days of the proestrous-estrous interval. In dogs subjected to laparoscopy, initial evidence of the presence of visible preovulatory follicles was closely related to the first detectable elevations in both estrone and estradiol-1 713. Either estrone or estradiol-1 713 concentrations or both surged sharply in 24 of the 25 cycles examined; however, only titers of estradiol-17j3 were elevated above baseline in all cycles prior to, or at the time of, the LH peak. In 16 of 25 cycles, estradiol-1713 concentrations declined by 20 pg/mI or more 12-24 h prior to the onset of sexual receptivity. Bitches remained in estrus during periods of declining estrone and estradiol-1 713and increasing progesterone levels. These data integrate the hormonal, ovarian and behavioral events of the estrous cycle of the bitch and suggest that: 1) a preovulatory estradiol-17(3 surge exists and is likely responsible for triggering LH release; 2) estrone may play a supportive role to estradiol-1713 in the endocrine control of LH secretion; 3) preovulatory changes in follicular morphology are distinct and can be correlated with a shift from estrone and estradiol-17f3 to progesterone secretion; 4) prolonged sexual receptivity in this species is supported in the presence of declining estrogen and continuously rising progesterone concentrations.

Abstract: Selective adherence to host mucosal surfaces is probably a requirement for colonization and infection by bacteria. Since pharyngeal colonization may be an important determinant in the pathogenesis of pneumonia, we studied the adherence of 10 different bacteria to pharyngeal cells obtained from nonsmokers, smokers, and chronic bronchitics. Various patterns of adherence among the different groups of subjects were found. Young healthy smokers had increased adherence of Streptococcus pneumoniae type I and, to a lesser extent, S. pneumoniae type III and Staphylococcus aureus when compared with nonsmokers. Middle-aged smokers with a long history of chronic bronchitis had significantly increased adherence only of untypable Haemophilus influenzae when compared with age-matched nonsmokers. The acquisition of pneumococcal pneumonia by smokers and the role of nontypable Haemophilus species in chronic bronchitis may be determined, in part, by bacterial adherence to pharyngeal cells.

Abstract: Two cases of small cell tumor of the uterine cervix are reported in which electron microscopic studies indicated that the tumor cells were of neural crest origin. Both tumors behaved aggressively, and metastatic involvement of pancreas was demonstrated in one patient, while the second patient developed liver metastasis. The need to distinguish these neoplasms from squamous cell carcinoma of the cervix is stressed. The tumors are compared with small cell neuroepithelial neoplasms previously reported as adult neuroblastomas. Cancer 43:1138–1145, 1979.

Abstract: Nine men with chronic erectile dysfunction (three primary, six secondary) who had no regular sexual partner were treated in two 12-session all-male psychoeducational therapy groups. Treatment interventions addressed specific factors which inhibited adequate sexual function with a focus on coping skills to overcome those factors. Pre, post, and follow-up behavioral self-report data and responses on a goal attainment scale questionnaire indicated that the treatment groups were successful for five men with secondary and one man with primary erectile dysfunction. Subjective report and pre- and posttreatment fantasy productions to TAT cards for the first group indicated that all men significantly improved their attitudes about sexuality and their sexual self-concept. The results suggest that this is a viable, cost-effective treatment for secondary erectile dysfunction, but not for primary erectile dysfunction unless supplementary individual therapy is provided.

Abstract: A second major species of leucine tRNA, tRNA Leu UAG (formerly designated tRNA Leu CUA) was purified from baker's yeast in a three-step procedure entailing BD-cellulose chromatography in the presence and absence of Mg2+ and Sephadex G-100 gel filtration. Results of aminoacylation and partial RNase T1 digestion experiments showed that this tRNA retains a native conformation under conditions that denature yeast tRNA Leu m5CAA (tRNA3 Leu). The primary structure of baker's yeast tRNA Leu UAG was elucidated by application of sensitive radioactive isotope derivative ("postlabeling") methods. Complete RNase T1 and A and partial RNase U2 fragments, prepared from non-radioactive tRNA and 5'-half and 3'-half molecules, were separated by two-dimensional polyethyleneimine-cellulose anion-exchange thin-layer chromatography and isolated by a novel micropreparative procedure affording high yields of these compounds in sufficient purity for subsequent tritium derivative analysis. Base composition and sequence of oligonucleotides were analyzed by tritium derivative methods. Molar ratios of the fragments were determined from the radioactivity of 3H-labeled nucleoside trialcohols in combination with base analysis. 2'-O-Methylated guanosine was characterized using the [gamma-32P]ATP/polynucleotide kinase reaction. The analysis of classical complete and partial RNase digests by the tritium derivative methods yielded the complete nucleotide sequence of the tRNA. A total of about 20 A260 units of the RNA was used for analysis, i.e. considerably less material than required for conventional spectrophotometric analysis. A different sequencing approach, consisting of a combination of "readout sequencing" with tritium sequencing of complete RNase T1 and A fragments, was applied to the 3'-half molecule. The 3'-half molecule was labeled with 32P at its 5' terminus, partially degraded with RNase T1, U2, and Phy1 and with alkali, and subjected to polyacrylamide gel electrophoresis. The sequence was read off the gel on the basis of cleavage patterns and size of the fragments. While the readout procedure provided only the positions of A, U, C, and G residues in the chain, additional information from tritium derivative analysis was utilized to define the positions of the modified nucleosides. The readout sequencing procedure was found to require less than 0.01 A260 unit of RNA and the analysis of the complete fragments about 6 A260 units. Interesting structural features of tRNA Leu UAG are (a) the location of unique, leucine tRNA iso-acceptor-specific sequences next to U-8, a constant nucleotide participating in synthetase recognition, (b) the occurrence of 1-methyladenosine in the T loop, a modification not present in the structurally related tRNA Leu m5CAA, and (c) the unusual presence of an unmodified uridine in the first position of the anticodon, which may be related to the unusual coding properties reported for this tRNA.

Abstract: The role of dying cells in the optic stalk in relation to retinal fiber migration was investigated in the chick embryo. Cell death was analysed at various stages of development by counting pycnotic nuclei and also by the Gomori acid phosphatase reaction, while nerve fibers were visualised by the Bodian method. A wave of cell death, beginning in the neural retina at stage 18 and advancing with time through the stalk towards the diencephalon, occurred simultaneously or slightly prior to differentiation and migration of ganglion cell axons. Cell death stopped and gliogenesis occurred in the stalk after penetration by retinal fibers. Cell death occurred in the stalk even when fiber penetration was prevented by optic cup ablation. In this case, necrosis ensued until almost complete degeneration of the stalk, usually within three days after the operation, and gliogenesis did not occur. As the stalk degenerated, its cells became heavily pigmented. These observations suggest that the onset of cell death in the optic stalk is determined prior to and independently of retinal fiber penetration. On the other hand, cessation of cell death and subsequent gliogenesis occur only in the presence of ingrowing optic fibers.

No factors were identified that influenced survival. Thromboembolic episodes and second primary tumors commonly complicate the clinical course. Metastases in widespread organs are common autopsy findings. Immunological mechanisms may explain these findings.

Abstract: Myoclonus occurs in a variety of pathological conditions, some inherited. We recently evaluated 3 members of a Louisiana-Texas family with an autosomal dominant disorder manifested by adult-onset, generalized, stimulus-sensitive myoclonus and slowly progressive distal muscle weakness and wasting. The analyses of cerebrospinal fluid homovanillic acid and 5-hydroxyindoleacetic acid before and after probenecid provided some evidence of impaired turnover of central dopamine and serotonin. Treatment with clonazepam resulted in complete and lasting improvement of the myoclonus. A postmortem examination in 1 member of the family revealed chiefly neuronal degeneration of the anterior horn cells, Clark's nucleus, and the lower cranial nerve nuclei. A similar syndrome has not previously been reported.

Abstract: Cervical radiculopathy unaccompanied by pain or sensory disorder but manifested only by paresis, atrophy, fasciculation, and reflex loss is unusual. Three such cases are presented. Considerable diagnostic difficulty may arise in distinguishing patients presenting with these features from those who have primary motor neuron disease. Two additional case reports demonstrate that this distinction is not always possible. Diagnosis, management, and prognosis are discussed. Differentiation between motor neuron disease and spondylosis will avert needless surgery in the former group and will result in beneficial, sometimes curative surgery in the latter group.

Abstract: The time course of appearance of hyperplastic alveolar nodule(s) (HAN) and mammary tumor(s) (MT) was determined in untreated virgin "viable yellow" (Avy/A) and non-yellow (A/a) (C3H/HeNIcrWf X VY/Wf)F1 female mice. The first HAN was detected in a yellow female at age 16 weeks, the first period in which mice were killed. HAN were not found in the non-yellow mice until they were 19 weeks old. The incidence of HAN increased to 92% among yellow females and to 75% among non-yellow females by 36 weeks of age. MT were first observed at age 22 weeks in yellow mice and at 28 weeks in non-yellow mice. The incidence of MT at 36 weeks was 75% among yellow mice and 22% among non-yellow mice. Type B adenocarcinoma was the predominant class of MT found in the yellow mice. The time courses of appearance and incidence of HAN and MT in the non-yellow F1 mice were similar to those observed in inbred C3H female mice. MT first appeared in each population when the incidence of HAN bearers had reached 40--45% regardless of age, body weight, or number of HAN per HAN bearer. Apparently, the phenotypic effects of the Avy gene primarily stimulated the multiplication of nodule-transformed cells to form HAN and thus indirectly enhanced MT formation.

Abstract: SummaryAn aerosol generator suitable for administration of a small-particle aerosol of amantadine for treatment of influenza A infection in man is described. As currently operated, the usual daily recommended oral dose of amantadine of 120 to 200 mg can be given in 8 to 10 hr of inhalation of the aerosol. Limited clinical study indicates the safety and probable efficacy of this method of treatment.
