Abstract: The effects of high-intensity pulsed electric fields (HIPEF) treatment (35 kV/cm, 4 μs bipolar pulses at 200 Hz for 800 or 1400 μs) on the microbial stability, quality parameters and antioxidant properties of a fruit juice–soymilk (FJ–SM) beverage along the storage time at 4 °C were compared to those obtained by thermal pasteurization (90 °C, 60 s). HIPEF processing for 800 μs ensured the microbial stability of the beverage during 31 days; however, longer microbial shelf-life (56 days) was achieved by increasing the treatment time to 1400 μs or by applying a thermal treatment. Peroxidase and lipoxygenase of HIPEF treated beverages were inactivated by 17.5–29% and 34–39%, respectively; whereas thermal treatment achieved 100% and 51%. During the storage, vitamin C content and antioxidant capacity depleted with time, and they were higher in FJ–SM beverages processed by HIPEF than in those thermally treated. Instead, total phenolic content of beverages did not present significant changes over the time, and it was higher in the 1400 μs-HIPEF treated beverages. In general, color, soluble solids, pH, and acidity values were not significantly affected by the processing treatments. Beverage viscosity increased over time, regardless of the treatment applied. Hence, application of HIPEF may be a good alternative treatment to thermal processing in order to ensure microbiological stability, high nutritional values and fresh-like characteristics of FJ–SM beverages.

Abstract: Aims: The objective of this study was to determine the temporal ochratoxin A (OTA) accumulation profile of Aspergillus section Nigri at different water activity (aw) levels. Methods and Results: Two Aspergillus carbonarius and two Aspergillus niger aggregate strains isolated from grapes were tested in vitro for OTA accumulation at 25°C on synthetic nutrient medium, over periods of 20 days at different aw levels. Results were modelled by a multiple linear regression and response surface predictive models were obtained. High levels of aw favoured OTA production by these moulds. Maximum amounts of OTA were found at the earlier growth states (5 days for A. carbonarius and 7–13 days for A. niger aggregate). Conclusions: Provided that A. section Nigri, and mainly A. carbonarius, play the main role in OTA presence in grapes, it would be critical to adjust the harvest and processing time to significantly reduce the chances for OTA accumulation. Significance and Impact of the Study: Ochratoxin A production by A. section Nigri has been shown for the first time to occur optimally after as little as 5 days on a grape-like medium.

Abstract: In semi-arid Mediterranean areas, studies of the performance of conservation tillage systems have largely demonstrated advantages in crop yield, soil water storage and soil protection against wind and water erosion. However, little attention has been given to interactions between soil biochemical properties under different tillage practices. Biochemical properties are useful tools to assess changes caused by different soil tillage systems in long-term field experiments. This study deals with the effect of long-term tillage practices (reduced tillage and no-tillage vs. traditional tillage) on soil chemical properties and microbial functions in three different sites of Spain (two of them located in the Northeast and one in the Southwest) under semi-arid Mediterranean conditions. Soil biological status, as index of soil quality, was evaluated by measuring microbial biomass carbon (MBC) and dehydrogenase (an oxidoreductase) and protease (a hydrolase) activities at three soil depths (0–5, 5–10 and 10–25 cm). In the three experimental areas, increases in soil organic matter content, MBC and enzymatic activities were found at the superficial layers of soil under conservation tillage (reduced tillage and no-tillage) in comparison with traditional tillage. Values of the stratification ratio of some biochemical properties were significantly correlated with yield production in Northeast sites. Conservation tillage has proven to be an effective strategy to improve soil quality and fertility in Mediterranean areas of Spain.

Abstract: Oxidative stress has been implicated in the pathogenesis of several neurodegenerative diseases including Alzheimer's disease (AD). Several proteins have been identified as targets of oxidative damage in AD dementia (usually stages V/VI of Braak) and in subjects with mild cognitive impairment associated with middle stages of AD pathology (stage IV of Braak). In this study, we investigate whether brain proteins are locally modified by oxidative stress at the first stages of AD-related pathology when morphological lesions are restricted to the entorhinal and transentorhinal cortices of neurofibrillary pathology (stages I/II of Braak). Using a proteomic approach, we show that the α subunit of the mitochondrial adenosine triphosphate (ATP)-synthase is distinctly lipoxidized in the entorhinal cortex at Braak stages I/II compared with age-matched controls. In addition, ATP-synthase activity is significantly lower in Braak stages I/II than age-matched control, while electron transport chain, expressed by the mitochondrial complex I activity, remains not affected. This is the first study showing oxidative damage in the first stage, and clinically silent period, of AD-related pathology characterized by entorhinal and transentorhinal tauopathy.
