Abstract: X-linked adrenoleukodystrophy is a neurometabolic disorder caused by inactivation of the peroxisomal ABCD1 transporter of very long-chain fatty acids. In mice, ABCD1 loss causes late onset axonal degeneration in the spinal cord in association with locomotor disability resembling the most common phenotype in patients, adrenomyeloneuropathy. Increasing evidence indicates that oxidative stress and bioenergetic failure play major roles in the pathogenesis of X-linked adrenoleukodystrophy. In this study, we aimed to evaluate whether mitochondrial biogenesis is affected in X-linked adrenoleukodystrophy. We demonstrated that Abcd1 null mice show reduced mitochondrial DNA concomitant with downregulation of mitochondrial biogenesis pathway driven by PGC-1α/PPARγ and reduced expression of mitochondrial proteins cytochrome c, NDUFB8 and VDAC. Moreover, we show that the oral administration of pioglitazone, an agonist of PPARγ, restored mitochondrial content and expression of master regulators of biogenesis, neutralized oxidative damage to proteins and DNA, and reversed bioenergetic failure in terms of ATP levels, NAD+/NADH ratios, pyruvate kinase and glutathione reductase activities. Most importantly, the treatment halted locomotor disability and axonal damage in X-linked adrenoleukodystrophy mice. These results lend support to the use of pioglitazone in clinical trials with patients with adrenomyeloneuropathy and reveal novel molecular mechanisms of action of pioglitazone in neurodegeneration. Future studies should address the effects of this anti-diabetic drug on other axonopathies in which oxidative stress and mitochondrial dysfunction are contributing factors.

Abstract: This report reviews the clinical applications of routine ultrasound in gestation control programmes for dairy herds. Emphasis is placed on identifying risks of late embryo/early foetal loss of non-infectious cause by monitoring ovarian structures (as opposed to uterine contents), indicators of pregnancy loss and twin pregnancies. Suggestions are provided to appropriately manage a cow at risk of pregnancy loss during the late embryonic/early foetal period. This review identifies gaps in our current knowledge of the aetiology of pregnancy loss and spontaneous embryo/corpus luteum reduction. Two areas of the implantation-placentation window of gestation in particular need urgent attention: the roles in pregnancy loss of immune-endocrine interactions and the physiological status of the embryo/corpus luteum. The latter issue and consecutively endocrine interactions could be successfully addressed by incorporating Doppler ultrasonography into routine ultrasound procedures.

Abstract: Small portable rainfall simulators are considered a useful tool to analyze soil erosion processes in cultivated lands. European research groups in Spain (Valencia, Malaga, Lleida, Madrid and La Rioja), France (Reims) and Germany (Trier) have used different rainfall simulators (varying in drop size distribution and fall velocities, kinetic energy, plot forms and sizes, and field of application) to study soil loss, surface flow, runoff and infiltration coefficients in different experimental plots (Valencia, Montes de Malaga, Penedes, Campo Real and La Rioja in Spain, Champagne in France and Mosel-Ruwer valley in Germany). The measurements and experiments developed by these research teams give an overview of the variety of methodologies used in rainfall simulations to study the problem of soil erosion and describe the erosion features in different climatic environments, management practices and soil types. The aims of this study are: (i) to investigate where, how and why researchers from different wine-growing regions applied rainfall simulations with successful results as a tool to measure soil erosion processes; (ii) to make a qualitative comparison about the general soil erosion processes in European terroirs; (iii) to demonstrate the importance of the development of standard method for measurement of soil erosion processes in vineyards, using rainfall simulators; and (iv) and to analyze the key factors that should be taken into account to carry out rainfall simulations. The rainfall simulations in all cases allowed infiltration capacity, susceptibility of the soil to detachment and generation of sediment loads to runoff to be determined. Despite using small plots, the experiments were useful to analyze the influence of soil cover to reduce soil erosion, to make comparisons between different locations, and to evaluate the influence of different soil characteristics. The comparative analysis of the studies performed in different study areas points out the need to define an operational methodology to carry out rainfall simulations, which allows us to obtain representative and comparable results and to avoid errors in the interpretation in order to achieve comparable information about runoff and soil loss.
