A rapid and efficient procedure for the purification of DNA from agarose gels.

TLDR
DNA fragments electrophoresed through a horizontal agarose slab gel can be recovered by inserting strips of filter paper backed by dialysis membrane into slits cut in the gel in front of the DNA bands and continuing Electrophoresis until the DNA is collected in the paper.

Studies on the transformation of intact yeast cells by the LiAc/SS‐DNA/PEG procedure

TL;DR: The LiAc/SS‐DNA/PEG method was shown to be more effective than other treatments known to make cells transformable and caused tighter binding of 32P‐labelled plasmid DNA than did double‐stranded (DS) carrier.
Journal ArticleDOI

A reliable method for the recovery of DNA fragments from agarose and acrylamide gels

TL;DR: The method is useful for the recovery of biologically active DNA from both agarose and high-percentage polyacrylamide gels, thereby enabling purification of DNA fragments up to 20 kb in size with a yield of 60–80%.
Journal ArticleDOI

Pichia pastoris as a host system for transformations.

TL;DR: A methylotrophic yeast, Pichia pastoris, is developed as a host for DNA transformations based on an auxotrophic mutant host of P. pastoris which is defective in histidinol dehydrogenase.
Journal ArticleDOI

Applications of high efficiency lithium acetate transformation of intact yeast cells using single-stranded nucleic acids as carrier.

TL;DR: Co‐transformation showed that 30–40% of the transformation‐competent cells take up more than one DNA molecule which can be used to enrich for integration and delection events 30‐ to 60‐fold.
Journal ArticleDOI

Expression of the lacZ gene from two methanol-regulated promoters in Pichia pastoris

TL;DR: Two DNA fragments containing putative control regions regulating the expression of the alcohol oxidase and dihydroxy-acetone synthase genes from the methylotrophic yeast Pichia pastoris were used in the construction of vectors for theexpression of the Escherichia coli lacZ gene.

A new technique for the assay of infectivity of human adenovirus 5 DNA.

TL;DR: A new technique for assaying infectivity of adenovirus 5 DNA has been developed and a reproducible relationship between amounts of DNA inoculated per culture and numbers of plaques produced was demonstrated.

A method for the recovery of DNA from agarose gels

H.F. Tabak, +1 more
TL;DR: A quick and versatile method for the isolation of DNA from agarose gels by putting the hydroxyapatite on a small column of Sephadex G50, elution and subsequent removal of phosphate can be performed in one step.
