TLDR
There have been mixed results from large adjuvant metastasis-prevention studies of bisphosphonates, but there are strong signals from large subgroups analyses of randomised phase III trials suggesting significant anti-tumour beneficial effects in specific patient populations.

Crosstalk between bone niche and immune system: Osteoimmunology signaling as a potential target for cancer treatment

TL;DR: The co-regulation of the skeletal and immune systems strikingly exemplifies the extreme complexity of such an interaction and their interdependency must be considered in designing therapeutic approaches for either of the two systems.

Incidence, risk factors and prognostic characteristics of bone metastases and skeletal-related events (SREs) in breast cancer patients: A systematic review of the real world data.

TL;DR: The unique characteristics of BM and SREs in breast cancer patients should be taken into account in future randomized controlled trials, as to optimize individual treatment options and assure a maximally long good quality of life.

Differences of osteoblastic bone metastases and osteolytic bone metastases in clinical features and molecular characteristics.

TL;DR: Understanding the differences of bone metastases between two phenotypes is important to help clinicians to understand the underlying mechanisms, behaviors and therapies in development and currently available forBone metastases.

Update on breast cancer risk prediction and prevention.

TL;DR: It still remains unclear how to prevent oestrogen-negative breast cancer, which occurs more often in premenopausal women, and the inclusion of breast density and a panel of genetic polymorphisms, which individually have low penetrance, can improve risk assessment.

Metastasis to bone: causes, consequences and therapeutic opportunities

TL;DR: The most common human cancers — lung, breast and prostate — have a great avidity for bone, leading to painful and untreatable consequences.

Nitrogen-Containing Bisphosphonates Inhibit the Mevalonate Pathway and Prevent Post-Translational Prenylation of GTP-Binding Proteins, Including Ras

TL;DR: Nitrogen‐containing bisphosphonate drugs cause apoptosis following inhibition of post‐translational prenylation of proteins such as Ras, and the data support the view that clodronate acts by a different mechanism.

Cellular and molecular mechanisms of action of bisphosphonates

TL;DR: Bisphosphonates currently are the most important class of antiresorptive agents used in the treatment of metabolic bone diseases, including tumor‐associated osteolysis and hypercalcemia, Paget's disease, and osteoporosis.
