Evidence for a thyroid peroxidase associated “active iodine” species

TLDR
Results support the idea that thyroid peroxidase plays a direct catalytic role in iodination, which does not occur by way of free iodine generated by the enzyme and released into the medium, but rather via an iodinating species associated with the enzyme.

A stereochemical concept for the catalytic mechanism of prolylhydroxylase: applicability to classification and design of inhibitors.

TL;DR: The Stereochemical mechanism presented here allows a detailed understanding of the kinetic findings and of the electronic rearrangements during the reaction, and invites pinpoint alterations of the enzyme substrates, leading to the design of structures with inhibitory potential.
Journal ArticleDOI

Structural and functional aspects of thyroid peroxidase.

TL;DR: The latest contributions in the field of TPO research are reviewed and a large reference list of original publications is provided to investigate the mechanisms whereby TPO contributes to hormone synthesis and constitutes an important autoantigen involved in autoimmune thyroid disease.
Journal ArticleDOI

The irreversible inactivation of thyroid peroxidase by methylmercaptoimidazole, thiouracil, and propylthiouracil in vitro and its relationship to in vivo findings.

TL;DR: The findings suggest that the initial in vivo action of these drugs is to block iodination by trapping oxidized iodide, not by acting as "general inhibitors" of the TPO.

The use of the 2-iminobiotin-avidin interaction for the selective retrieval of labeled plasma membrane components.

TL;DR: When intact human erythrocytes were sequentially labeled with periodate and 2-iminobiotin hydrazide and the Triton X-100-solubilized plasma membrane proteins were subjected to affinity isolation, the major sialoglycoproteins, periodic acid-Schiff (PAS) 1, PAS 2, and PAS 3, plus two proteins with apparent molecular weights higher than band 3 were retrieved.
Journal ArticleDOI

Reactions of purified hog thyroid peroxidase with H2O2, tyrosine, and methylmercaptoimidazole (goitrogen) in comparison with bovine lactoperoxidase.

TL;DR: The absorption spectra of thyroid peroxidase were similar to those of lactoperoxidases and intestinal peroxIDase, but obviously different from those of metmyoglobin, horseradish per oxidase, and chloroperoxidsase.

Disc electrophoresis-i background and theory*

TL;DR: Some mechanisms that provide a rationale for the resolution afforded by zone electrophoresis in many gels will be detailed; the theory of some new modifications of zone electophoresis that have been designed to take maximum advantage of these mechanisms will be developed.

Peroxidase-Catalyzed Halogenation

TL;DR: A proposed mechanism for this reaction suggests that the oxidation is mediated by the iodoenzyme derivative followed by a prototropic rearrangement and scission to form the ether bound of thyronine and a serine residue on thyroglobulin.
