TLDR
The genomic sequencing procedures are applicable to the analysis of genetic polymorphisms, DNA methylation at deoxycytidines, and nucleic acid-protein interactions at single nucleotide resolution.Abstract:
Unique DNA sequences can be determined directly from mouse genomic DNA. A denaturing gel separates by size mixtures of unlabeled DNA fragments from complete restriction and partial chemical cleavages of the entire genome. These lanes of DNA are transferred and UV-crosslinked to nylon membranes. Hybridization with a short 32P-labeled single-stranded probe produces the image of a DNA sequence "ladder" extending from the 3' or 5' end of one restriction site in the genome. Numerous different sequences can be obtained from a single membrane by reprobing. Each band in these sequences represents 3 fg of DNA complementary to the probe. Sequence data from mouse immunoglobulin heavy chain genes from several cell types are presented. The genomic sequencing procedures are applicable to the analysis of genetic polymorphisms, DNA methylation at deoxycytidines, and nucleic acid-protein interactions at single nucleotide resolution.read more

Efficient in vitro synthesis of biologically active RNA and RNA hybridization probes from plasmids containing a bacteriophage SP6 promoter
TL;DR: In this paper, a simple and efficient method for synthesizing pure single stranded RNAs of virtually any structure is described, based on the unusually specific RNA synthesis by bacteriophage SP6 RNA polymerase which initiates transcription exclusively at an SP6 promoter.

TL;DR: A genomic sequencing method is reported that provides positive identification of 5-methylcytosine residues and yields strand-specific sequences of individual molecules in genomic DNA, which suggests that the high methylation level of single-copy sequences in sperm may be locally modulated by binding of protein factors in germ-line cells.

TL;DR: A gene discovered by positional cloning has been identified as the von Hippel-Lindau (VHL) disease tumor suppressor gene, and a restriction fragment encompassing the gene showed rearrangements in 28 of 221 VHL kindreds.

The beige mutation in the mouse selectively impairs natural killer cell function.
TL;DR: It is reported here that a mutant gene in the mouse called beige (bgJ), leads to a complete and selective impairment of naturally occurring killer lymphocytes, whereas all other forms of cell-mediated lysis are apparently normal.

Antigenic modulation of Bordetella pertussis.
TL;DR: The antigenic structure of Bordetella pertussis varies continuously with environmental conditions of growth and, under suitable conditions, any of three forms (or modes) with markedly different antigenic surfaces may be developed.

TL;DR: The results of this study indicate that scid specifically impairs the differentiation of stem cells into mature lymphocytes; myeloid cell differentiation is not affected.
